MT2A基因多态性与2型糖尿病的关系

Association between MT2A gene polymorphismand type2 diabetes mellitus

摘要: 目的:探讨16号染色体长臂(16q)金属硫蛋白(metallothionein,MT)基因多态性与中国北方汉族人群2型糖尿病的关系。方法:采用PCR技术和限制性片段长度多态性(RFLP)的方法检测129名糖尿病患者(病例组)和115名正常对照者(对照组)的MT基因家族中的MT2A基因rs10636位点单核苷酸多态性。结果:rs10636 3种基因型GG、GC和CC的频数分布符合Hardy-Weinberg平衡;MT2A的3种基因型GG、GC和CC的频数分布在病例组和对照组中无统计学意义(χ2=0.202,P>0.05),等位基因G和C的频数分布在病例组和对照组中也无统计学意义(χ2=0.136,P>0.05)。结论:MT2A基因rs10636位点可能与2型糖尿病无关。

Abstract: Objective To investigate the genetic association of metallothionein (MT) 2A gene locus in chromosome 16 with susceptibility to type 2 diabetes mellitus (T2DM) in Chinese Han people. Methods The techniques of PCR and restriction fragment polymorphism(RFLP)were used to examine single nucleotide polymorphism(SNP)of MT2A gene in 129 patients with T2DM(case group) and 115 unrelated healthy individuals(control group). Results The distributions of genotype frequencies of rs10636 were not deviated from the Hardy-Weinberg equilibrium; there were no remarkable differences of the frequency distributions of GG,GC and CC of MT2A between cases and controls (χ2=0.202,P>0.05),and the frequency distributions of G and C had no markedly differences between cases and controls (χ2=0.136,P>0.05).Conclusion The rs10636 of MT2A gene may not be associated with T2DM.
