survivin在喉癌组织中表达及其与细胞凋亡的关系

Expression of survivin gene in laryngeal carcinoma and its relationship with apoptosis

摘要: 目的:探讨凋亡抑制蛋白survivin在喉癌中的表达及其与凋亡和临床病理参数的关系。方法:应用免疫组织化学方法检测喉癌细胞株Hep2、牙周膜细胞及40例喉癌组织和8例正常喉黏膜中survivin蛋白的表达,半定量逆转录PCR检测survivin mRNA表达,DNA原位末端标记技术(tunel)检测喉癌组织的细胞凋亡指数(AI)。结果:survivin在正常喉黏膜、牙周膜细胞中不表达,Hep2细胞株和57.5%(23例)喉癌组织表达survivin蛋白和mRNA。在病理学分级为Ⅰ级的喉鳞癌标本中,survivin蛋白阳性率(33.3%)明显低于Ⅱ、Ⅲ级的标本(72.7%,P<0.05);临床Ⅲ~Ⅳ期喉鳞癌患者survivin蛋白阳性表达率为76.2%,明显高于临床Ⅰ~Ⅱ期患者(36.8%, P<0.05);喉癌颈部淋巴结转移组中survivin 蛋白阳性率(100%)显著高于未转移组(48.5%,P<0.05)。survivin表达阳性组的AI 为2.93±1.20,阴性组为4.05±1.42,两者比较,差异具有显著性(P<0.05)。结论:survivin在喉癌中高表达可能通过抑制细胞凋亡而在喉癌的发生发展中发挥一定作用。

Abstract: Objective To study the expression of a novel apoptosis inhibitor,survivin,in laryngeal carcinoma and its relationships with apoptosis and clinical pathologic parameters. Methods The expressions of survivin protein in Hep2 cell line, periodontal ligament cells (PDLC),40 laryngeal carcinoma and 8 normal laryngeal tissue were detected by immunohistochemical staining. The expression of survivin mRNA was detected by RT-PCR. DNA in situ end labelling technique was used to detect apoptosis index (AI). Results Survivin protein expressed in Hep2 cell line and 23 of 40 (57.5%) cases of laryngeal carcinoma. In contrast, it didn′t express in PDLC and normal laryngeal tissues. The positive percentage of survivin protein in tumor grade Ⅰ laryngeal carcinoma (33.3%) was strikingly lower than that of grade Ⅱ and Ⅲ laryngeal carcinoma (72.7%,P<0.05). The positive percentage of survivin protein in clinical stage Ⅲ and Ⅳ laryngeal carcinoma (76.2%) was strikingly higher than that of grade Ⅰand Ⅱ laryngeal carcinoma (36.8%, P<0.05). The positive percentage of survivin protein in laryngeal carcinoma with lymph node metastasis (100%) was strikingly higher than that of laryngeal carcinomas without lymph node metastasis (48.5%,P<0.05,). AI of survivin protein positive cases (2.93±1.20) was lower than that of negative cases(4.05±1.42,P<0.05). Conclusion The results indicate that apoptosis inhibition caused by survivin protein may play a important role in the origin and progression of laryngeal carcinoma.
