Role of basolateral amygdala in startle conditioned reflex

摘要: 目的:验证杏仁基底外侧核(BLA)在惊恐条件反射中的作用。 方法:随机将大鼠分成2组。急性实验组(n=35)大鼠麻醉后,用玻璃微电极观察和记录其BLA神经元的自发放电及电击足底和声音刺激引起的诱发反应;慢性实验组(n=25)大鼠在麻醉状态下做股动脉插管手术,术后恢复3 d,开始进行惊恐条件反射训练。该训练是将条件刺激(声音刺激)和非条件刺激(足部电击)相结合,训练后,大鼠对单独的条件刺激产生惊恐反应。此时用0.3 μL 2%利多卡因阻断BLA,观察大鼠对条件刺激产生惊恐反应的改变。 结果:急性实验组大鼠,电击足底和声音刺激可引起BLA一些神经元放电频率较刺激前明显增加(放电频率变化超过20%);慢性实验组大鼠,训练前,单独给予声音刺激不引起血压发生明显改变(P>0.05);训练后(需经3 d),单独给予声音刺激可引起血压明显升高(P<0.01)。此时,用利多卡因阻断BLA的作用,再单独给予声音刺激不再引起血压明显升高(P>0.05)。 结论:BLA神经元可感受声音和电击足底两种不同的刺激;同时,经惊恐条件反射训练的大鼠对单独的声音刺激产生的惊恐反应需要BLA的参与。

Abstract: Objective To study the role of basolateral amygdala (BLA) in startle conditioned reflex. Methods The rats were divided into two groups. The responses of neuronal discharges of BLA to the foot shock and the tone stimulus by glass microelectrodes were observed and recorded in acutely experimental rats. The cannulation of femoral artery in chronically experimental rats was performed. The startle conditioned reflex was started after 3 days. The training was to combine a conditioned stimulus (a tone) with an unconditioned stimulus (a foot shock), the startle responses were established by only conditioned stimulus in chronically experimental rats. After the roles of BLA were blocked by 0.3 μL 2% lidocaine, the response to conditioned stimulus was observed. Results The neuronal discharges of BLA were increased after the tone stimulus or the foot shock (change of discharges frequency>20%) in the acutely experimental rats. The blood pressure was not increased before training (P>0.05), but the blood pressure was increased after 3 days training (P<0.01) by only tone stimulus in the chronically experimental rats. The blood pressure was not increased by only tone stimulus (P>0.05) when the BLA was blocked by lidocaine. Conclusion The BLA neurons can experience the tone and the foot shock and play an important role in startle conditioned reflex.
