硒和维生素E对心肌成纤维细胞Ⅰ型胶原合成代谢的影响

Effects of Se and vitamin E on anabolism of type Ⅰcollagen in cultured cardiac fibroblasts

摘要: 目的:探讨硒(Se)和维生素E(VE)对心肌成纤维细胞Ⅰ型胶原合成代谢的影响。方法:用不同浓度的Se和(或)VE作用于体外培养的Wistar大乳鼠心肌成纤维细胞,测定培养上清液中羟脯氨酸及Ⅰ型胶原的含量。结果:①低、中和高浓度Se(0.05、0.1和0.2 mg·L-1)均可抑制NE(0.2 mg·L-1)的促成纤维细胞(CFbs)胶原合成的作用(P<0.05),以高浓度Se最为明显(56.13±1.31,P<0.05);②不同浓度VE(1、10和100 mg·L-1)均降低NE对CFbs胶原合成的促进作用,以1和100 mg·L-1 的VE对羟脯氨酸含量的影响最显著(P<0.05);③Se和VE的联合应用(0.05 mg·L-1 Se+10 mg·L-1 VE),对CFbs的胶原合成的抑制作用更明显(41.34±1.73,0.39±0.04,P<0.05)。结论:Se和VE对心肌成纤维细胞Ⅰ型胶原合成代谢有抑制作用。

Abstract: Objective To inform the effects of Se and vitamin E on the anabolism of type Ⅰ collagen in cultured cardiac fibroblasts. Methods Cardiac fibroblasts were cultured using culture solution containing different concentrations of Se and vitamin E (VE), Se+VE for 48 h, then the contents of hydroxyproline and type Ⅰ collagen in cultured solution were detected. Results ①Compared with control and NE groups, the contents of hydroxyproline could be significantly decresed in all the three Se group (low 0.05 mg·L-1, middle 0.1 mg·L-1, high 0.2 mg·L-1) (P<0.05), and this effect became apparent when the content of Se was 0.2 mg·L-1 (56.13±1.31, P<0.05); ②Compared with control and NE groups, the contents of hydroxyproline were decreased significantly in all the three VE groups (1, 10, and 100 mg·L-1), especially in 1 and 100 mg·L-1 groups(P<0.05); ③Both the contents of hydroxyproline and type Ⅰ collagen were decreased when Se (0.05 mg·L-1) and VE (10 mg·L-1) applied simultaneously. Conclusion The anabolism of type Ⅰ collagen in cultured cardiac fibroblasts can be inhibited by Se and VE.
