铜绿假单胞菌普通菌毛与耐药性R质粒的关系

Relationship between pili and resistance R plasmidin strain PA36 of P.aeruginosa

摘要: 目的:检测铜绿假单胞菌耐药株PA36的耐药性R质粒与菌毛及粘附性的关系。 方法:应用琼脂糖凝胶电泳试验、电镜照片制备及观察、粘附试验及质粒消除试验。 结果:PA36含有一条相对分子质量是3.3×106的R质粒;电镜下观察PA36具有周毛型菌毛;粘附试验于光镜下可见该菌大量粘附到脱落的尿道上皮细胞上;该R质粒消除后该菌株的耐药性与菌毛一起消失,粘附性明显减弱。 结论:铜绿假单胞菌菌株PA36编码普通菌毛、决定粘附性的基因主要是在相对分子质量为3.3×106的一个R质粒上。

Abstract: Objective To study the relationship between the resistance R plasmid and both pili and adherence of Pseudomonas aeruginosa strain,PA36. Methods Agarose gel electrophoresis of plasmid DNA, adherence test, and extract of plasmid were used. Electron microscope photos were prepared and observed. Results The wild type strain of PA36 contained a plasmid (3.3×106), heavy pili were found on the surface of PA36 under electron microscope, crowed bacteria cells adhered to dropped urethra-epithelial cells under light microscope. After the plasmid of PA36 was cured by SDS, both the resistance and pili almost completely disappeared and the capability of adherence weakened sharply in the meantime. Conclusion The results suggested that gene encoding ordinary pili of wild type PA36 may be located in the R plasmid, which is responsible for the adherence of PA36. Thus, the R plasmid in PA36 also names adherence plasmid. The gene also is relates to the pili and the capability of adherence.
