金属硫蛋白基因多态性与2型糖尿病的关系

Association between metallothionein gene polymorphismand type 2 diabetes mellitus

摘要: 目的:探讨金属硫蛋白(metallothionein,MT)基因多态性与中国汉族人群中2型糖尿病(T2DM)发病之间的关系。 方法:采用PCR技术和限制性片段长度多态性(RFLP)的方法检测41名糖尿病患者和53名正常对照者的金属硫蛋白基因家族中的MT4基因(rs666636)201G→A突变,统计各组对象的突变频率。 结果:MT4基因(rs666636)201G→A突变型等位基因(A)频率在糖尿病组和正常对照组中差异有显著性(P<0.05,OR=2.335);AA, GA和GG三种基因型频率分布差异也有显著性 (P<0.05);不同基因型的相对风险分析, GA基因型携带者患2型糖尿病的风险是GG基因型的3.462倍(OR=3.162,95% OR CI=1.389~8.629)。 结论:MT4突变型等位基因与2型糖尿病的发生有显著的关联性,基因突变使2型糖尿病的发病风险增加。

Abstract: Objective To detect the association between metallothionein (MT) gene polymorphism and type 2 diabetes mellitus (T2DM) in Chinese Han people. Methods The techniques of PCR and restriction fragment length polymorphism (RFLP) were used to examine MT4 gene (rs666636) G→A mutation in 41 patients with T2DM and 53 unrelated healthy individuals, and then the mutation frequency was statistically computed. Results The alleic frequency (A) of MT4 gene mutation was significantly different between the diabetic and control group (P<0.05,OR=2.335).The distribution rates of AA, GA and GG had also remarkable differences (P<0.05).The relative risk of suffering from T2DM of GA genotype was 3.462 times (OR=3.462,95% OR CI=1.389~8.629) GG genotype. Conclusion There′s an association between the alleic gene of MT4 gene mutation and T2DM, and the mutation genotype has increased the risk of suffering from T2DM.
