Changes of IL-8 and IL-10 in lung tissues with hyperoxia-induced injury

Abstract:

Abstract:Objective To study the changes of interleukin 8 (IL-8) and interleukin 10 (IL-10) in hyperoxia-induced lung injury and significances. Methods  The model of rat with hyperoxia-induced lung injury was established by being inhaled with high concentration of oxygen (>95%).The lung tissues were harvested 6 h and 1,3,6,10,14 d  after oxygen treatment, the histopathological changes of lung tissues  were observed by HE staining and Double-antigen sandwich ELISA was used to detect the IL-8 and IL-10 levels in the lung tissue homogenate. Results  The infiltrative inflammatory cells appeared in alveolus first,and then alveolus was enlarged ,then alveolar septum became thinner and eventually the number of alveolus was decreased as the time of exposure to oxygen prolonged. The concentrations of IL-8 in lung tissue homogenate in high  oxygen group were significantly higher than those in control group, the level reached the peak at  day 3 after high oxygen exposure and the statistical difference lasted until the 14th day. The concentrations of IL-10 in lung tissue homogenate were significantly higher after high oxygen exposure 6 h and also had statistical difference on day 1, but restored normal level at day 3. Conclusion The concentration of inflammatory factor IL-8 is increased fast and lasts for long time while anti-inflammatory factor IL-10 is increased for short time in hyperoxia-induced lung injury.
