Effects of traditional puffball on proliferation and collagen synthesis of skin fibroblasts in rats

Abstract:

Abstract:Objective To study the effects of puffball on proliferation and collagen synthesis on rat skin fibroblasts, and to investigate the  mechanism of  enhancing wound healing  of  active constituent of puffball     at the cells level.Methods The rat skin fibroblasts were divided into 5 groups, all the cells were treated with puffball(37.5,75.0,150.0 and 300.0 mg/L), except for control group,for 24,48 and 72 h,respectively.And then MTT was used to detect the proliferation, and the content of collagen were measured by double antibody sandwich ELISA.Results  Compared with control group, the cells treated with different concentrations (37.5,75.0,150.0 and300.0 mg/L ) of puffball showed a significant rise both in its proliferation and the ability of collagen synthesis (P<0.01).Furthermore, in time-dependant manner,  the proliferation rate of cells and collagen synthesis ability  wer  significantly increased after treated for 24,48,72 h,especially in 72 h(P<0.01). Conclusion  Puffball with different doses  can enhance the proliferation and collagen synthesis in  skin fibroblasts of rats.
