Abstract:

Abstract:Objective To investigate the effect of fluoxetine on the expression of 5-HT type 1A (5-HT1A) receptor in hippocampus tissue of posttraumatic stress disorder (PTSD) rats and to explore the mechanism of fluoxetine in the treatment of PTSD.Methods The male Wistar rats were randomly divided into control group, model group and fluoxetine group. The rat PTSD model was set up using SPS & S method.Hippocampus tissue was isolated and the expression of 5-HT1A receptor wasdetected with Western blotting and RT-PCR.Results The 5-HT1A receptor protein expressions in hippocampus tissue of rats in control group, model group, and flouxetine group were 1.18±0.12,0.39±0.05,and 0.71±0.09,respectively. The 5-HT1A mRNA expressions were 0.95±0.12,0.19±0.05,and 0.74±0.05 in three group, respectively. There were significant differences between model group, flouxetine group and control group (P<0.05).Compared with model group, the 5-HT1A mRNA in flouxetine group was increased (P<0.05).The index was inhibited after treatment with furoxetin. Conclusion Fluoxetine could inhibit the 5-HT1A receptor expression in hippocampus of PTSD rats.
