Establishment and evaluation of determination of astragaloside Ⅳ in Baoyuanqingxue granules by HPLC

Abstract:

Objective To examine the various factors that affect determination of astragaloside Ⅳ in Baoyuanqingxue granules,elucidate the extraction method and chromatographic conditions of determination of astragaloside Ⅳ, and to establish an effective method for determination of astragaloside Ⅳ. Methods High performance liquid chromatography(HPLC) method was used.The SB-C18 column (Agilent,ZORBAX,4.6 mm×150 mm,5 μm)was adopted,the mobile phase was acetonitrile-warer(36∶64) at the flow rate of 1.00 mL?min-1 with UV detection at 203 nm,the volume of injection was 20 μL and the column temperature was 30℃. Results  The resolution between astragaloside Ⅳ and other peaks was good. The calibration curve was linear in the range of 0.010 03-0.200 60 g?L-1(r=0.999 9).The average recovery(n=6) of astragaloside Ⅳ was 98.4% with RSD of 0.8%. Conclusion  The HPLC-UV method to detect the cotent of astragaloside Ⅳ in Baoyuanqingxue granules is simple and accurate. It can be used for quality control of astragaloside Ⅳ in Baoyuanqingxue granules.
