Expressions of HBx and CEACAM1 in HBV-related hepatocellular carcinoma and their significances

Abstract:

Objective To investigate the expressions of hepatitis B virus X protein (HBx) and carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1) in HBV-related hepatocellular carcinoma tissue and to explore the relationship between the two genes and occurrence,development and invasion of hepatocelular carcinoma.
Methods  The expressions of HBx and CEACAM1 in 81 cases of HBV-related hepatocellular carcinoma tissues were detected by immunohistochemistry method,and the relationship with clinicopathological features was analyzed. The expressions of CEACAM1 in the human normal hepatocyte cell line QZG, hepatocellular carcinoma HepG2 and HepG2-X(HepG2 cells transfected with HBx) were detected by Western blotting. Results The positive rates of HBx and CEACAM1 expressions in 81 cases of HBV-related hepatocellular carcinoma tissues were 74.07%(60/81) and 71.60%(58/81), respectively. The expressions of HBx and CEACAM1 were correlated with portal vein invasion,nodal metastasis and TNM staging(P<0.05 or P<0.01). The expression of HBx was negatively correlated with CEACAM1(rs=-0.310,P<0.05).The CEACAM1 protein was significantly  decreased in HepG2-X compared with HepG2 and human normal hepatocyte cell line QZG.Conclusion The high expression of HBx and low expression of CEACAM1 are correlated with the invasion and metastasis of HBV-related hepatocelular carcinoma,and HBx may promote HBV related hepatocarcinogenesis and development by       inhibition CEACAM1.
