Construction of BMP7 eukaryotic expression recombination plasmid and its transfection in articular cartilages of rabbits

Objective To construct the eukaryotic expression vector of pcDNA3.1-BMP7 and transfect it into the articular chondrocytes of rabbit cultured in vitro in order to study the best condition of liposome and plasmid DNA. Methods Expression vector of pcDNA3.1-BMP7 was constructed and was digested by two enzymes and sequenced. pcDNA3.1-BMP7 was transfected into the articular chondrocytes of rabbit mediated with liposome and screened by G418.The best rates of liposome and G418, liposome and pcDNA3.1-B MP7 were identified. In situ hybridization, PCR and Western blotting were used to detect the expression of BMP7.Results The BMP7 gene of pcDNA3.1-BMP7 was same as the BMP7 gene reported.The best concentration was 10 μL liposome and 4 μg pcDNA3.1-BMP7 in 3 mL IMDM medium including 400 mg·L-1 G418. Conclusion Madiated with liposome reagent, the articular cartilages of rabbit are successfully transfected with pcDNA3.1-BMP7 and BMP7 express stably in the articular cartilages.

Association between KPNA1 gene polymorphism and schizophrenia

Objective To investigate the association between single nucleotide polymorphism of rs3762637 locus on KPNA1 gene and schizophrenia in the Northern Han Chinese. Methods The method of PCR-RFLP was conducted to examine the genotypes of KPNA1 gene in 101 core family (including patients and their parents ), and then the genotypic and allelic frequencies of KPNA1 gene were statistically computed.Results The χ2 goodness-fit test showed that the genotypic distributions of rs3762637 were not deviated from Hardy-Weinberg equilibrium either in the patient sample (χ2=1.431, df=1, P=0.232 ) or in the parent group ( χ2=1.212, df=1, P=0.271 ). There were no remarkable differences of the allelic and genotypic frequency distributions of KPNA1 between paren ts and patients( χ2=0.957, P=0.328;χ2=3.547, P=0.170 ). Transmission disequilibrium test ( TDT ) did not show significant biased transmission from parents to affected offspring (χ2=1.469, P=0.225). Conclusion The rs3762637 locus on KPNA1 gene may be not the susceptible locus of schizophrenia.

Repair of artificial epiphyseal plate cartilage on epiphyseal plate injury of rabbits

Objective To investigate the possibility of repairing the epiphyseal plate injury using artificial epiphyseal plate.Methods The left tibial epiphyseal plate of rabbit was resected about 1/3 to 1/2, then the artificial epiphyseal plate were transplanted; while the right side was injured but not proceed transplantation as contrast. The tibial angle was measured after X-ray check. T test was used for data analyzing. HE stain, toluidine blue Dyeing and immunohistochemistry test were performed to observe the histomorphology. Results: The tibial angle of lower extremity was (38.80±13.50)° in control group, (9.01±4.2)° in artificial epiphyseal plate group 8 weeks after transplantation; (54.25±18.33)° in control group, (11.27±5.1)° in artificial epiphyseal plate group 24 weeks after transplantation. There was significantly difference between control group and artificial epiphyseal plate group (P<0.01). There was bony bridge formation in control group. 2 weeks after planting, the epiphyseal plate impairment region was filled with cartilage tissue, but the chondrocytes in the transplanted epiphyseal plate were changed into irregular arrangement, and demarcation was distinct with normal epiphyseal plate cartilage. After planting for 4 weeks, cartilage layers were noted in the treatment groups and the cutting edges disappeared. There were collagen-Ⅱ-positive cells in transplanted epiphyseal plate cartilage.Conclusion The artificial epiphyseal plate can repair the epiphyseal plate injury, and the effect is satisfied.

Objective To express and secrete the natural N-terminal rBPTI with PHOⅠ signal peptide in Pichia pastoris. Methods PHOⅠ/bpti genes were inserted into the eukaryotic expression plasmid. The recombinant plasmid was transformed into the Pichia pastoris (X-33) via electroporation. Results The expression plasmid was constructed to contain correct sequence for PHOⅠ/bpti genes. The rBPTI was expressed and secreted in X-33. An activity strain was selected with trypsin inhibitor experiment .The expression supernatant was purified with cation exchange chromatography to single peak and SDS-PAGE indicated that the relative molecular mass was 6 500. Conclusion rBPTI with natural N-terminal sequence is successfully expressed in Pichia pastoris with signal peptide of PHOⅠ.

Objective To explore the effect of apoptosis on chondrocyte differentiation. Methods The apoptosis of costal chondrocytes of 5-week-old and 12-week-old rabbits was detected by methods of transmission electron microscopy and TUNEL. Results The proliferation and differentiation of chondrocytes was a gradual procedure. The apoptotic chondrocytes occurred at boundary from proliferation zone to hypertrophy zone by using transmission electron microscopy. The apoptosis existed in the whole cartilage zone, but the apoptotic percentage was highest in hypertrophy zone and increased with the age by using TUNEL method. Conclusion The proliferation and differentiation of costal chondrocytes from silence zone to bone zone of rabbits is a gradual procedure, in which apoptosis may play an important regulatory effect.

Objective To study the induction of islet-like cell from bone marrow mesenchymal stem cells (MSCs) in vitro and its repair effects on injury pancreas tissues.Methods ①MSCs were cultivated and expanded in vitro and in duced with cell factors like bFGF, EGF, high glucose DMEM media into islet-like cells. The induced cells were identified by dithizon staining. ②MSCs and bone marrow cells were marked with DAPI and implanted into experimental group (MSCs implantation) and control group (bone marrow mononuclear leukocyte implantation) . The survival rate and morphological changes were observed two weeks after implantation. Results ①After the amplified MSCs were induced in vitro,their forms changed from long spinde-shape to polygon or round and attached each other. Dithizon staining showed bright red color for most of induced cells. It indicated MSCs could be induced into islet-like cell s. ②The survival rate of the experimental group was 80%. The injured pancreas tissues were repaired.Blue fluorescent was observed in repaired pancreas tissues. The animals in control group all died during two weeks. Conclusion MSCs can be induced into islet-like cells in vitro and repair the injured pancreas.

Objective To discuss the effects of different intensity of HVEF on physiological function of animal organism.Methods 150 Kunming mice were randomly divided into five groups with 30 animals in each group: normal control group, four experimental groups treated with 0.6, 1.2, 1.8 and 2.4 kV·cm-1 HVEF, respectively. The variation of physiological and biochemistry indexes in the developmental pr ocess of mice which underwent different intensity of HVEF were measured. Results HVEF with a certain intensity could promote mouse development, the threshold value was 0.6 kV·cm-1 . Treated with this intensity,HVEF could increase the weight of mice and the activities of SOD and GSH-Px in both cardiac muscle cells and hepatocytes, and decreased the content of LPO, compared with control group, the difference was significant (P<0.01). HVEF pro moted the metabolism by regulating the activities of various enzymes. Conclusion HVEF can improve mouse development not only by single effect on some metabolism mechanism, but also by the coordination of the whole organism,the suitable HVEF is of benefit to rehabilitation process of many kinds of diseases.

Objective To examine the expression and distribution of six transmembrane epithelial antigen of the prostate (STEAP1) in human prostate tissues. Methods The RT-PCR for mRNA, Western blotting for protein, immunohistochemistry for distribution were carried out. Results The results of RT-PCR and Western blotting showed that STEAP1 mRNA and protein highly expressed in prostate cancer tissues than those in normal prostate tissues. Immunohistochemistry staining showed that most staining positive cells were prostate epithelial cells , they located in the membrane of epithelial cells, and mainly in the side of cell membrane to the glandular vessels of the prostate.Conclusion STEAP1 express widely in human prostate cancer tissu es, and is closely related to epithelial cells, which is closely connected with the glandular of prostate. The expression and distribution indicate that STEAP1 is involved in substance metabolism and transportation in prostate.

Action of lowering blood sugar levels of corn active polysaccharide on diabetic mice

Objective To study the action of corn active polysaccharide (CAP) on blood sugar levels in diabetic mice.Methods Fifty mice were randomly divided into control group, alloxan model group, CAP 3.50,1.75, 0.88 g·kg-1 groups (n=10). Mice in each group were exhibited with CAP once a day for 15 d,on the sixth day,mice in every group were injected with alloxan physiological salt solution 80 mg·kg-1 in caudal vena except control group, mice in control group were injected with physiological salt solution at the same volume. 16 h before the last exhibition, mice in every group were starved. 1 h before the last exhibition, blood was extracted in eyeballs, sera were separated and the contents of blood sugar were determined by auto-biochemistry analyzer, mice were killed at once. The liver samples were obtained and the contents of liver glycogen were determined. Another 60 mice were divided into 5 groups with the method men tioned above. Each group was exhibited once a day for 10 d continuously. 1 h after the last exhibition, except control group, the mice were injected with adrenalin hydrochloride 250 μg·kg-1 through abdominal cavity, after30 min, blood was extracted by decapitation, and the contents of blood sugar and liver glycogen were determined by auto-biochemistry analyzer.Results CAP 3.50 g·kg-1 had obvious inhibitory effect on the raise of blood sugar in diabetic mice (model group)(P<0.05), it had obvious action on reducing the contents of liver glycogen in diabetes mice(model group)(P<0.05),and 3.50 g·kg-1 was superior to 1.75 g·kg-1. CAP with different doses had obvious actions on inhibiting the raise of blood sugar in adrenine mice(model group)(P<0.001, P<0.01, P<0.05),and had obvious action on increasing the reduction of liver glycogen on adrenine mice(model group),and 3.50 g·kg-1 was superior to 1.75 g·kg-1. Conclusion CAP has the function of lowering blood sugar levels in alloxan diabetic mice and hyperglycemia mice induced by adrenine.

Objective To investigate the effects of EGF and collagen scaffold on cultured rat dermal fibroblasts in vitro. Methods The fibroblasts were isolated by enzyme digestion and cultured with DMEM medium contained new bovine serum (10%). The proliferation abilities of the fibroblasts were evaluated by MTT assay and the morphologic changes of the fibroblasts were evaluated by histological method.Results When the EGF concentration was above 5 μg•L-1, the proliferation abilities of cultured fibroblasts increased markedly. The number of cultured fibroblasts planted into collagen scaffold increased slowly. Conclusion EGF has the auxoaction on proliferation of the cultured fibroblasts in vitro. Collagen scaffold has some regulatory effects on proliferation rate of the cultured fibroblasts in vitro.

Objective To observe the distribution and change regularity of molybdenum (Mo) and copper (Cu) in blood and tissues in rabbits fed on supplement with different levels of Mo,and to study the biological antagonism of Mo on Cu.Methods Fifty rabbits were divided into five groups:control group and four experimental groups (treated with different doses of Mo). The experimental time was four months. The contents of Mo and Cu in tissues of the rabbits were determined. Results Both Mo and Cu contents were increased in blood,liver,kidney,lung,spleen,pancreas,myocardium and muscle of rabbits with the increase of Mo intake in the diet(P<0.01). The contents of Mo and Cu in kidney were increased remarkably, and closely related to Mo intake (r=0.9995,r=0.9997). The contents of Mo in bone, cartilage and hair were also increased (P<0.01), but not Cu when compared with control group. Conclusion The antagonism of Mo on Cu may be formed. Because Mo and Cu may be related to the stable Mo-Cu-protein complex without biological activity in blood or tissues. The complex heaping in blood or tissues can not be taken good advantage by the organism so that the animals show symptom of cooper-deficiency.

Objective To investigate the analgesic effect of saponin of Pterocarpus Indicus(SPI). Methods Sixty rats were divided randomly into 5 groups (n=12):SPI with doses of 15,30,60 μg·kg-1 groups,SPI plus naloxone group and control group. Through the cannula penetrated into lateral cerebral ventricle, SPI was injected into lateral cerebral ventricle. Then,the tail-flick latency after the injection of SPI naloxone and saline was measured. Results The tail-flick latency was increased significantly 5 min after the injection(i.c.v.) of SPI,the amplitudes were corresponding with their doses. The effect of increasing of pain threshold in 30 μg·kg-1 SPI group was maintained until 50 min after the injection of SPI, 60 μg·kg-1SPI group was maintained until 60 min after the injection of SPI(P<0.01 or P<0.01).The pain threshold in control group was unchanged in 60 min (P>0.05). Naloxone could effectively antagonize the enhancement of the pain thresho ld induced by SPI.Conclusion SPI has a strong analgesic action and shows an obvious dose-effect relationship. The analgesic action may be mediated by μ opioid receptor in brain.

Objective To establish and evaluate epileptic model kindled by kainic acid microinjected into nucleus amygdalae. Methods Kainic acid was microinjected (1 μg) into right nucleu s amygdalae of Wistar male rats with stereotaxic technique. Praxiology and elect rophysiology observation were performed after kainic acid injection, as well as pathological detection on 6th, 12th, 72th hour and 7th, 14th, 21st day, respec tively. Results The experimental rats presented typical epileptic seizures relapse and various epileptic discharge in parietal cortex electroenciphalogram. The nerve cell damage was observed through HE staining in hippocampus or temporal lobe. The kindling rate of the model was 90%.Conclusion The epileptic model has been successfully established by kainic acid microinjected into nucleus amygdalae in Wistar male rats, this model is applicable for temporal lobeepilepsy.

Differentiation of human umbilical cord blood mononuclearcells into hepatocyte-like cells induced in vitro

Objective To investigate whether or not human umbilical cord blood mononuclear cells (MNC) could be induced to differentiate into hepatocyte-like cells in vitro.Methods Human umbilical cord blood samples were obtained sterilely by punture umbilical vein, MNC were isolated by lymphocyte separation medium (density 1.077 g·mL-1). The surface antigen expressions of MNC were detected by flow cytometry. MNC were induced with DMEM medium, containing HGF alone, FGF4 alone, HGF+FGF4 or no growth factor, respectively. The markers of hepatic linear cells were examined before induction, and 7, 14, 21 and 28 d after induction by RT-PCR, immunocytochemistry and periodic acid-schiff (PAS) in every group. Results C-met and FGFR2 mRNA were detected before induction. AFP and Albumin mRNA were detected on 7th and 21th day after induction.CK18,hepat ocyte antigen,glycogen were detected on 28th day after induction . The markers of hepatocyte linear cells did not appear in no growth factor induction group.The positive percents of every hepatocytic marker at the same time point had statistically differences between HGF+FGF4 and HGF alone or FGF4 alone (P<0.05).Conclusion Both HGF and FGF4 can induce umbilical cord blood MNC to differentiate into hepatocyte-like cells singly or coordinately. 

Nephritic gene expression of scavenger receptor A in adriamycin nephrosis rats

Objective To study the relationships between nephritic gene expression of scavenger receptor A, lipid metabolism and pathology in adriamycin nephrosis rats.Methods Injecting of adriamycin in veins of the rat′s tail was used to make adriamycin nephrosis model, PCR and RT-PCR were used to measure the gene expression levels of scavenger receptor A in different groups. Results Compared with control group, protein excretory quantity in adriamycin nephrosis group was significantly increased (P<0.05),the levels of serum total protein and albumin were remarkedly decreased (P<0.05), while the levels of TG, TC, HDL, LDL, ApoA, ApoB were increased (P<0.05) . The gene expression value of scavenger receptor A in adriamycin nephrosis group (0.74±0.14) was higher than that in control group (1.17±0.13) (P<0.01). Glomerulus epithelium foot appeared confluence, endodermis cells proliferated, in some serious ones the basis membrane of glomerulus was unclear, endodermis cell was archoid observed under electron microscope in adriamycin nephrosis rats. Conclusion The gene expression value of scavenger receptor A is lower in normal rats, and the high value of scavenger receptor A is closely related to abnormal of lipid metabolism in adriamycin nephrosis rats. It is also one of the main reasons of pathologic changes of the kidney.

Experimental study on small gaps bridging and epineurialsuture in repairing rupture of peripheral nerves

Objective To discuss the curative effects of small gaps bridging and epineurial suture in repairing the rupture of peripheral nerves. Methods One side of sciatic nerve of 60 SD rats was treated by small gaps briding(experimental group), another side was treated by epineurial suture (control group). Results In experimental group, the recovery time of autonomous motion (40 d) was shorter; nerves growed normally, nerve trunk was intact,the nerve conduction velocity was fast when observed under microscope;the proliferation of connective tissues in small gaps decreased markedly, nerve fiber arranged regularly when observed with histological method;there was no difference between regenerative nerves in the distal gaps and normal nerves when observed under transmission electron microscope. And there were no changes mentioned above in contro l group.Conclusion This method of small gasps bridging can repair the rupture of nerves in great degree,and its curative effect is superior to that of epineurinal suture.

Objective To screen the incidence of mtDNA 7445G point mutation among the patients with sensorineural hearing loss (SNHL). Methods COI gene mutations were detected in 150 cases of SNHL (aged from 5 to 19 years old) with the method of PCR-RFLP-DNA sequencing.Results COI gene 7445 point mutation was not found in the all samples with SNHL. One novel 7444A point mutation was observed in one case of SNHL.Conclusion The 7445G mutation is seldomly appealed among the cases of SNHL in China, 7444A mutation may be one of the molecular biological basics related to the aminoglycoside antibiotic-induced deafness.

Objective To study the protective effects of fenugreek seeds components A, B on renal lesion in diabetic rats.Methods The model of diabetes mellitus was built with streptozotocin (STZ) in rats. All the rats were randomly divided into four groups: diabetes control group (D) (n=8), fenugreek seeds components A treatment group (A) (n=10), fenugreek seeds components B treatment group (B) (n=10), and normal control group (C) (n=6). Diabetic rats were treated with fenugreek seeds components A,B for 12 weeks, The metabolism levels of blood glucose, blood lipids were determined. Kidney morphology was observed to evaluate the protective effects of fenugreek seeds components A, B on renal structure and function in diabetic rats.Results In diabetic rats treated with fenugreek seeds component s A and B , the levels of blood glucose, urinary protein excretion, serum creatinine, blood urea nitrogen, kidney index and glomerular volume were significantly lower than those in group D (P>0.05,P<0.01). In group A, the levels of TG, LDLC were lighter than those in group D (P>0.05), HDLC was higher than that in group D (P<0.01),CHO had no significant difference with group D. In group B, the levels of TG, LDLC and CHO were lighter than those in group D, whereas the level of HDLC was higher than that in group D (P<0.01). Optical and electromicroscopic findings indicated that the lesions of glomerular of rats in group B were alleviated significantly, and the glomerular structure in group B was better than that in group A. Conclusion Fenugreek seeds components A, B can modulate the metabolism of blood glucose, blood lipids in diabetic rats. Functional and morphological abnormalities in kidney have been lightened. Fenugreek seeds component s may protect the kidney of diabetic rats to a certain degree. The protective effect of components B is better than that of components A.

Objective To study the protective effects of flavone mixture of crataegus leaves(FMCL) on cerebral ischemia. Methods The local cerebral ischemia model was initiated by the middle cerebral artery occlusion (MCAO) in rats for 4 h. The neurologic deficit score and the activities of creatine kinase (CK), lactata dehydrogenase (LDH), superoxide (SOD) and the content of serum malondialdehyde (MDA)in MCAO rats were measured. At the same time blood stasis models were built in rats. The effects of FMCL on platelet aggregation,blood viscosity (ηB) and plasma viscosity (ηP) were detected. Results Compared with the model group ,FMCL (60 and 30 mg·kg-1) increased SOD activity, decreased CK and LDH activities and serum MDA content in a dose dependent manner in MCAO rats (P<0.05). It could also decrease the neurologic deficit in MCAO rats (P<0.05) . And it could decrease platelet aggregation rate, ηB and ηP (P<0.01,P<0.05).Conclusion FMCL has protective effects on cerebral ischemia and its mechanism may be related to lowing ηB,ηP and preventing platelet aggregation.

Objective To investigate the preliminary effects and mechanism of oxymatrine(OMT) on analgesia. Methods Mice were randomly divided into 6 groups:normal saline group, OMT groups with different doses (50,100,and 200 mg·kg-1), pethidine hydrochloride (10 mg·kg-1)group, and NG-nitro-L-arginine methyl ester (L-NAME) 20 mg·kg-1+OMT 100 mg·kg-1 group. The number of writhing within 15 min was observed in pain mouse models caused by acetic acid. Femal e mice were randomly divided into 6 groups as the methods mentioned above. Micewere placed on the hot plate maintained at (55.0±0.5)°C and the latency of licking of the hind paws was observed. Results The number of writhing was decreased and the latency of licking of the hind paws was prolonged in a dose-pendent manner after intraperitoneal injection of OMT 50,100, and 200 mg·kg-1 in mice(P<0.05,P<0.01,P<0.001). L-NAME injected in advance enhanced the effect of OMT on antagonism of the writhing response caused by acetic acid.Conclusion These results suggest that OMT can antagonize the acute pain caused by acetic acid and hot plate in a dose-dependent manner in mice. The analgesic action of OMT may be not mediated completely by NO-cGMP pathway.

Objective To research the effect of Huangmo Polysaccharides extracts on liver cancer and its mechanism. Methods The bearing tumor mouse model with Hepatoma-22 was established for anti-tumor experiment. Huangmo Polysaccharides extracts (FI) was administered, i.p, its inhibitory rate of tumor (IR) was dtected. The lymphocyte transformation, the activity of NK cells, and the contents of IL-2 and TNF-α were determined with MTT assay.Results After treated with FI of 20,40, 80 mg·kg-1,the inhibition rates (IR) on Hepatoma-22 were 36.1%,47.7%,57.6%, respectively; It had a good activity on anti-liver cancer. FI increased the weights of immune organs, improved the lymphocyte transformation and the activities of NK cells, Il-2 and TNF-α. Conclusion As biological response modifier, FI has effects of anti-liver cancer through increasing the activity of body immunity, improving the lymphocyte transformation and the activities of NK cells, Il-2 and TNF-α.

Objective To explore the changes of serum TNF-α and IL-6 contents in the two-hit rat models with hemorrhage and lipopolysaech (LPS) and the effects of panaxadiol saponins (PDS) on TNF-α and contents IL-6. Methods Adult Wistar rats were randomly divided into 5 groups: sham operational group (S), hemorrhage group (H), two-hit group with hemorrhage and LPS groups (HL), Dexamethasone pretreatment group (HLD), PDS pretreatment group (HLP). The rat models were made by hemorrhagic shock as the first hit and with endotoxin as the second hit. Then the rats were killed after 6 h. The contents of serum TNF-α and IL-6 in rats were measured by radioimmunoassay.Results The serum TNF-α and IL-6 contents in HL group were increased significantly compared with S group or H group (P<0.001). The TNF-α and IL-6 contents in HLP group and HLD group were significantly lower than those in HL group (P<0.01). Conclusion LPS can increase significantly the contents of serum TNF-α and IL-6 in rats with hemorrhagic shock. PDS can inhibit the relea se of serum TNF-α and IL-6, and has the same effects with DEX to protect against the tissue injuries of two-hit rats with hemorrhage and LPS.

Objective To study the establishment and stability of hepatic cirrhosis animal model(CM). Methods The methods of compound factor, carbon tetrachloride(CCl4) plus alcohol, corn flour and cholesterol plus alcohol were used to construct the hepatic cirrhosis rat models. 80 Wistar rats were selected, male and female were ,respectively. There were 40 Wistar rats treated with compound factor method.The fodder feed was 80% corn flour,20% animal oil, and 0.5% cholesterol, the only beverage was 5% alcohol .At the first time,the Wistar rats were administered with 40% CCl4-olive oil mixture by subcutaneous injection (5 mL·kg-1). Then they were hypodermically injected with 40% CCl4-olive oil mixture (3 mL·kg-1) every 3 days for six weeks. At the end of the 6th week,six Wistar rats were executed randomly and made into hepatic smears which were accorded with hepatic cirrhosis (The forming of pseudolobuli was the only standard to decide the successful establishment of CM). The rest rats were randomly divided into two groups. There were 13 Wistar rats treated with normal fodder feeding in each group. Meanwhile the rats in one group were hypodermically injected with 40% CCl4-olive oil mixture (3 mL·kg-1) every 7 days. Then the pathological changes at the end of 7th and 8th week were observed. Twenty Wistar rats treated with CCl4 plus alcohol were fed with normal fodder food. Remain ing conditions were as same as those were used in the first 6 weeks of compound factor method. Twenty Wistar rats treated with corn flour and cholesterol plus alcohol weren′t hypodermically injected with CCl4 and remaining conditions were same with those were used in the first 6 weeks of compound factor method.Results The rat models with hepatic cirrhosis treated with compound factor were established completely at the end of 6th week. Then the rats were fed with normal fodder food. Pseudolobuli were all disappeared after two weeks. And the rats were hypodermically injected with CCl4 for 2 weeks after establishment of CM,pseudolobuli were mainly existed. Typical CM were not constructed with other two methods. Conclusion The forming rate of hepatic cirrhosis model constructed with compound factor is high. After establishment of the model, CCl4 can keep the relative stability of the models.

Methods The different volume fractions of Zirconia nano-powder were added into micrometer Alumina powder and the mechanical properties of the samples were tested. Results The sample with 10%(V/V)ZrO2(3Y) nano powder had the best mechanical properties,the flexural strength and fructure toughness were (424.43±30.30)MPa and(4.31±0.30)MPa m1/2,respectively. Conclusion The mechanical property of nano-ceramic composite with 10%(V/V)ZrO2(3Y) nano powder could fit the demand of prothodontics,excessive content of nano-powder decrease the samples′ properties on the contrary.

Objective To study the effects of Shugan Jieyuling self-made (SJSM) on behavior, levels of serum IL-2, IL-6 and cortisol in depression model mice. Methods The adult mice separately raised and treated with chron ic unpredictable middle stress stimulus were used to establish depression models. The curative effect of SJSM was observed in depression model mice.The changes of weight and behavior were detected in a period. Radioimmunoassay(RIA) was used to examine the contents of serum IL-2, IL-6 and cortisol levels. Results The increased weights of the depression model mice were declined compared with the normal mice before administration. The mental state and behavior of the depression mice were changed. The mice were starling, dreadful, helpless and immobile. At the same time the contents of serum IL-2, IL-6 and cortisol were obviously lower than those of the normal mice. SJSM (large and low doses) and Baiyoujie changed the increased weights and behaviors of the depression after administration for 21 d. The mental state was meliorated simultaneously,and the serum IL-2, IL-6 and cortisol levels in the depression model mice were decreased significantly compared with normal mice. Conclusion The levels of serum IL-2, IL-6 and cortisol may be the guideline for the diagnosis of depression disease. SJSM can obviously improve both the symptoms of the depression models and the levels of serum IL-2, IL-6 and cortisol.

Objective To observe the effects of puerarin on the morphology,cell proliferation activity,and cell relative survival rate of Chinese hamster lung cells(CHL). Methods MTT colorimetric assay was used to detect the effects of puerarin with different concentrations at different time on the survival rate of the CHL. Results The survival rates of CHL were obviously decreased under the exposure of various concentrations (12.50,50.00,100.00,200.00,400.00 mg·L-1) of puerarin,proliferation of CHL was significantly inhibited and some cells detached from the wall in puerarin groups and presented a dose-time dependent manner, the 50% inhibited concentraion (IC50)for cell growth was 282 mg·L-1.  Conclusion Puerarin might inhibit the proliferation and the metabolic activity of mitochondria of CHL.

Objective To determine the effects of bisphosphonate (YM175) on the quality of bone morphogenetic protein (BMP)-induced bone. Methods Bisphosphonate was administered to rats in which subcutaneous ectopic bones were induced by recombinant human BMP-2. Bisphosphonate (YM175) 1 μg·kg-1·d-1 was administered to rats carrying the BMP-induced bones three times per week, from the 3rd to 7th week after BMP implantation (bisphosphonate group). Aliquots of phosphate buffered saline were administered in the same protocol without bisphosphonate in the control group. At the 3rd, 4th, 7th or 10th week, the BMP-induced bones were removed and observed by contact microradiography(CMR) and hematoxylin and eosin(H&E)staining. Results By 3 weeks when bisphosphonate started to be administered, woven bones formed in the periphery of the BMP pellets. At the 4th week , in both the bisphosphonate and control groups, bone formation advanced inward. During the 10 weeks, bone marrow was formed and the characteristics of lamellar bones, in which the lanunae of the small osteocytes were regularly arranged, were noted in the control group. Conclusion These findings suggest that bone resorption can be inhibited by the bisphosphonate.

Objective To study the changes of different mass-ratio of polylactic acid/glycolicacid (PLGA) common bile duct stent in bile in vitro, including mass, relative molecular mass as well as physical appearance in order to find an ideal substitute for T tube in common bile duct exploration.Methods 45 stents of four different ratios (50/50, 60/40, 70/30 and 80/20) of PLGA were used respectively. All the stents were submerged into the vials with fresh human bile and incubated at 37℃. The bile was changed every day and the changes of mass, relative molecular mass and physical appearance were observed every other day. Results The degradation peak times of 50/50 and 60/40 ratio was 9-10 d, that of 70/30 stents was 13-14 d, and 80/20 ratio was 17-18 d. The degradation peak time of 80/20 ratio was consistent with the termination time(14 d ) of T tube.Conclusion The degradation time of 80/20 ratio stents is consistent with the termination time of T tube, T tube can be replaced by this ratio stent.

Objective To investigate the functions of tumor necrosis factor-alpha (TNFα) and tumor necrosis factor receptor (p55 TNFR) on the process of normal pregnancy and the pathogenesis in pregnancy-induced hypertension syndrome (PIH). Methods Placental tissues were collected with cesarean-section in 12 normal pregnant women and 43 PIH women. TNFα and p55 TNFR expressions in placental tissues were detected by immunohistochemical technique. Results The results demonstrated that TNFα and p55 TNFR were expressed in placental tissues in both normal pregnant and PIH groups. The staining degrees in patients with moderate and severe PIH were higher than those in the groups of normal pregnancy and mild PIH (P>0.05). In severe and moderate PIH groups, the staining degrees of maternal side of placental tissue were higher than that of fetal side (P<0.01).Conclusion TNF-α and p55 TNFR expressions in small scale in n ormal pregnant women may participate to regulate the normal pregnancy, whereas the over expression of TNF-α and p55 TNFR in placenta may play an important role in the pathogenesis of PIH.

Objective To explore the effects of XIAP on carcinogenesis and progression of breast cancer.Methods The expressions of XIAP in the tissues of normal mammary (96 cases), cystic hyperplasia mammary (56 cases), atypical hyperplasia mammary (12 cases),and breast cancer (119 cases) were evaluated by SP immunohistochemistry. The relationships between XIAP expression and the pathologically biological features of breast cancer were assessed.Results The positive rates of XIAP in the tissues of normal mam mary, cystic hyperplasia mammary, atypical hyperplasia mammary and breast cancer were 4.2%(4/96), 5.4%(3/56),42.7%(5/12) and 72.3%(86/119), respectively. The positive rates in the last two groups were higher than those in the former two groups (P<0.05). XIAP was more expressed in the infiltrative non-special breast cancer (82.0%,73/89) than in the special and early stage infiltrative breast cancer (37.5%,3/8)(P<0.05). Expression of XIAP was increased in breast cancer with lymph node metastasis although the difference was not significant (P>0.05). Conclusion Overexpression of XIAP is common in carcinogenesis and progression of breast cancer. Altered expression of XIAP may contribute to carcinogenesis and progression of breast cancer by inhibiting apoptosis, its overexpression indicates worse prognosis.

Objective To explore the histopathological and immunohistochemical features of gastrointestinal stromal tumor(GIST). Methods Histopathological and immunohistochemical detections were peformed in 22 cases of primary GIST with CD117, CD34, Desmin,SMA, S-100 protein by SP methods. Results The primary cell morphology consisted of spindle shaped cells and epithelioid cells. Cystification, mucification, hemorrhage or necrosis presented at the same time. The mitotic count was higher than 5/50HPF in 5 cases in which the diameters of tumor were above or equal to 5 cm. Cystification, hemorrage or necrosis were found in 4 cases and mucification in 1 case. The mitotic count was 2-5/50HPF in 2 cases which were epithelioid cell and tumor diameter were 2-4 cm with mucification in 1 case. CD117 and CD34 showed diffuse strong expression with immunohistochemical staining, the posit ive rates were 95.5% and 68.2%, respectively. Desmin almost never presented, S-100 protein and SMA focally presented or were absent. Conclusion The morphology of GIST consist of various kinds of cell types and different arrangements. Immunohistochemical staining is positive for CD117 and CD34, negative for desmin, focally positive or absent for S-100 protein and SMA. The tumor′s diameters are above or equalto 5 cm, mitotic count is over 5/50HPF, hemorrhage or necrosis might be helpful to identify tumors with malignant potential.

Objective To study the relations between the expression of P16 in gastric carcinoma (GC) tissues and the clinical characteristics of GC, then discuss the effect of P16 on genesis and development of GC. Methods Expressions of P16 were examined by S-P immunohist ochemica1 staining in 10 cases of normal gastric tissue (NGT) and 63 cases of GC . Results ①The positive rate of P16 expression in NGT was 80.0%, the positive rate in GC was 44.4%. There was significant difference between them (P<0.05). ②There were no significant differences of the positive rate of p16 expression between cardiac part,fundus and body of stomach, and pyloric part (P>0.05). ③There was significant difference of the positive rate of P16 expression between cases with high differentiation and moderate-low different iation gastric carcinoma (P<0.05). ④Between cases with invasion through muscle and not, there was significant difference (P<0.05). ⑤Significant difference could be found between cases with lymph node metastasis and not (P<0.05). Conclusion There are closely relations between the expression loss of P16 and gastric carcinoma. The P16 expression imperfection is indiscriminate in GC which developed under different invasion factors.

Objective To investigate the statistical association between the G/C promoter polymorphism at (-174) of the IL-6 gene and transient ischemia attack(TIA) events, as well as the progress of cerebral infarction in 3 months.Methods 121 TIA patients and 114 age- and sex- matched healthy control subjects were examined for the (-174) IL-6 G/C polymorphism by mutagenic separated polymerase chain reaction (MS -PCR).The progress of cerebral infarction in 3 months was recorded. Results In the patients with TIA,the pro-portion of genotype GG(54.6%)was significantly higher than that in the control group(38.6%)(P<0.05),and also higher than those of GC(30.6%) and CC (14.9%)(P<0.05).The incidence rate of cerebral infarction in 3 months had significant differences between GG patients(28/66,54.6%) and GC(6/37,16.2%),CC(3/18, 16.7%)(P<0.05).Conclusion The genotype GG of IL-6 promoter is associated with pathogenesis and prognosis of TIA.

Objective To study the relationships of expressions of MMP-2, MMP-9, TIMP-1 in gastric carcinoma and pathological characteristics in clinic metastasis and prognosis. Methods Immunohistochemistry and morphologic quantitative analysis were used. Results Expression levels of MMP-2 and MMP-9 in gastric carcinoma matrix were markedly higher than those of pericarcinoma normal tissues (P<0.01). TIMP-1 expressed in pericarcinoma matrix. There were no any rel ationships between the expression levels of MMP-2, MMP-9, TIMP-1 in gastric carcinoma and patients′ age, tumor size, pathological grading, but there was obvious relationship between expression levels of MMP-2, MP-9, TIMP-1 and lymphnode metastasis (P<0.05). Conclusion There are close relationships between the expressions of MMP-2, MMP-9, TIMP-1 in gastric carcinoma and pathological characteristics in clinic metastasis and prognosis. They may be used as important indexes to judge the biological behavior and prognosis of gastric carcinoma.

Detection of IL-2 and T suppressor cell in patients with recurrent oral ulcer

Objective To investigate the immunologic mechanism of recurrent oral ulcer(ROU). Methods The IL-2 activities and T suppressor(Ts) cell functions in peripheral blood of 36 ROU patients were detected with CTLL and short-term Ts cell functional detection technique. Results The IL-2 activity of single-nuclei cells in peripheral blood of ROU patients was lower than that in normal controls (P<0.01), while the suppressing function of Ts cells had no difference between the two groups. The IL-2 activity of ROU patients increased sifgnificantly after treated with transfer factor compared with before treatment (P<0.05), while the Ts cell function did not change after treatment(P>0.05). Conclusion The ROU patients have a low cell immune function and an imbalanced T cell sub-clone, which may be related to the development of ROU. The effect of transfer factor on ROU is closely related to its function of up-reguating IL-2 activity.
