Protective effect of semen plantaginis extract on kidney of rats with membranous nephropathy and its mechanism

摘要: 目的

探讨车前子提取物(SPE)对膜性肾病大鼠肾脏的保护作用,并阐明其作用机制。

方法

将50只雄性SD大鼠随机分为对照组,模型组,低、中和高剂量SPE组,每组10只。除对照组外,其余各组大鼠采用阳离子化牛血清白蛋白诱导建立膜性肾病模型。造模成功后,低、中和高剂量SPE组大鼠分别灌胃给予剂量为1.5、3.0和4.5 mg·kg-1 SPE,模型组和对照组大鼠给予等量生理盐水,连续4周。生化法检测大鼠血清中肌酐(Scr)、尿素氮(BUN)和白蛋白(ALB)水平,TUNEL法检测各组大鼠肾组织细胞凋亡率,实时荧光定量PCR(RT-qPCR)法和Western blotting法检测各组大鼠肾组织中B细胞淋巴瘤-2(Bcl-2)、B细胞淋巴瘤-2相关蛋白X(Bax)、含半胱氨酸的天冬氨酸蛋白水解酶3(Caspase-3) mRNA和蛋白表达水平。

结果

与对照组比较,模型组大鼠血清中Scr和BUN水平明显升高(P<0.05),血清中ALB水平明显降低(P<0.05),肾组织中Bcl-2 mRNA和蛋白表达水平明显降低(P<0.05),肾组织中Bax、Caspase-3 mRNA和蛋白表达水平及肾组织细胞凋亡率明显升高(P<0.05)。与模型组比较,低、中和高剂量SPE组大鼠血清中Scr和BUN水平明显降低(P<0.05),血清中ALB水平明显升高(P<0.05),肾组织中Bcl-2 mRNA和蛋白表达水平明显升高(P<0.05),肾组织中Bax、Caspase-3 mRNA和蛋白表达水平及肾组织细胞凋亡率明显降低(P<0.05)。与低剂量SPE组比较,中和高剂量SPE组大鼠血清中Scr和BUN水平明显降低(P<0.05),血清中ALB水平明显升高(P<0.05),肾组织中Bcl-2 mRNA和蛋白表达水平明显升高(P<0.05),肾组织中Bax、Caspase-3 mRNA和蛋白表达水平及肾组织细胞凋亡率明显降低(P<0.05)。

结论

SPE通过调控Bcl-2和Bax mRNA及蛋白表达水平减少膜性肾病大鼠肾组织细胞凋亡,对膜性肾病模型大鼠肾脏起保护作用。

Abstract: Objective

To investigate the protective effect of semen plantaginis extract (SPE) on kidney of the rats with membranous nephropathy, and to elucidate its mechanism.

Methods

Fifty male SD rats were randomly divided into control group, model group, low, medium and high doses of SPE groups( n=10). In addition to control group, the rats in other groups were given cationic bovine serum albumin to establish the membranous nephropathy models. The rats in low, medium and high doses of SPE groups were given 1.5, 3.0 and 4.5 mg·kg-1 SPE by gavage, and the rats in model group and control group were given the same amount of normal saline, last for 4 weeks. The levels of creatinine (Scr), urea nitrogen (BUN) and albumin (ALB) in serum of the rats were determined. TUNEL assay was used to determine the apoptotic rates of kidney tissue cells of the rats in various groups. Real-time fluorescence quantitative PCR(RT-qPCR) and Western blotting methods were used to detect the expression levels of Bcl-2, Bax, Caspase-3 mRNA and proteins in kidney tissue.

Results

Compared with control group, the levels of serum Scr and BUN of the rats in model group were significantly increased (P<0.05), while the level of serum ALB was significantly decreased (P<0.05), the expression levels of Bcl-2 mRNA and proteins in kidney tissue of the rats were significantly decreased (P<0.05), the expression levels of Bax and Caspase-3 mRNA and proteins in kidney tissue were significantly increased(P<0.05), and the apoptotic rates were significantly increased (P<0.05). Compared with model group, the levels of serum Scr and BUN in low, medium and high doses of SPE groups were significantly decreased (P<0.05), while the level of serum ALB was significantly increased (P<0.05), the expression levels of Bcl-2 mRNA and protein and proteins in kidney tissue were significantly increased (P<0.05), the expression level of Bax and Caspase-3 mRNA and proteins in kidney tissue were significantly increased (P<0.05),and the apoptotic rates were significantly increased (P<0.05).Compared with low dose of SPE group, the levels of serum Scr and BUN in medium and high doses of SPE groups were significantly decreased (P<0.05),while the levels of serum ALB were significantly increased (P<0.05), the expression levels of Bcl-2 mRNA and protein in kidney tissue were significantly increased (P<0.05), the expression levels of Bax and Caspase-3 mRNA and proteins in kidney tissue were significantly decreased (P<0.05), and the apoptotic rates were significantly decreased (P<0.05).

Conclusion

SPE can protecte the kidney of the rats with membranous nephropathy by regulating the expression levels of Bcl-2 and Bax mRNA and proteins, and reduce the apoptosis of kidney tissue cells.
