四妙勇安汤对大鼠血栓闭塞性脉管炎的抗炎作用及其机制

Anti-inflammatory  effect of  Simiaoyong’an Decoction in thromboangiitis obliterans rats and its mechanism

摘要: 目的:观察四妙勇安汤(SMYA)对血栓闭塞性脉管炎(TAO)模型大鼠的治疗作用,并探讨其作用机制。方法:将实验大鼠随机分为假手术组、模型组、脉络宁阳性对照组(2.7 g/kg)及SMYA高、中、低剂量组(24.3、16.2和8.1 g/kg)。采用大鼠股动脉注射月桂酸钠法建立大鼠TAO模型,造模后灌胃给药,以脉络宁颗粒(MLN)为阳性对照药,连续给药15 d后处死动物取材,常规HE染色观察大鼠血管及周围组织的病理学变化,采用酶联免疫吸附法(ELISA)检测血浆中白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)、C-反应蛋白(CRP)、血栓素B2(TXB2)和6-酮-前列腺素F1α(6-K-PGF1α)含量,并应用紫外分光光度计测量血浆中一氧化氮(NO)的含量。结果:与模型组比较,SMYA高剂量组大鼠血浆中IL-6、TNF-α、TXB2和CRP含量明显降低,NO和6-K-PGF1α含量明显增加,差异均有统计学意义(P<0.05或P<0.01);SMYA中剂量组和低剂量组大鼠血浆中TNF-α含量明显降低(P<0.01)。模型组大鼠血管内皮受损严重,周围有大量炎细胞浸润;SMYA高剂量组大鼠血管皮细胞基本完整,有少量炎细胞浸润。结论:SMYA对TAO模型大鼠具有较好的治疗作用,其机可能与抗炎和维持血浆中血栓素A2(TXA2)和前列环素I2(PGI2)平衡功能有关。

Abstract: Abstract:Objective To observe the therapeutic effect of Simiaoyong’an Decoction (SMYA) on the rat models with thromboangiitis obliterans (TAO),and to explore the action mechanism.Methods The rats were randomly divided into 6 groups:sham operation group,model group,mailuoning group(MLN 2.7 g/kg),high,medium and low doses of SMYA groups (24.3,16.2,8.1 g/kg). The  TAO model was prepared by injecting sodium laurate into the femoral artery of rats.The rats were administered by intragastric administration after prepared model.After 15 d administration,the pathological changes of blood vessel and surrounding tissue were observed with HE staining,the contents of IL-6,TNF-α,CRP,TXB2 and 6-K-PGF1α in plasma were measured by enzyme-linked immunosorbent assay (ELISA),and the content of NO in plasma was measurd by ultraviolet spectrophotometer.Results Compared with model group,the contents of plasma IL-6,TNF-α,TXB2 and CRP in high dose of SMYA(24.3 g/kg) group were significantly decreased,and the contents of NO and 6-K-PGF1α were markedly increased (P<0.05 or P<0.01);the contents of plasma TNF-α in medium and low doses of SMYA (16.2 and 8.1 g/kg) groups were significantly decreased (P<0.01).The  vascular endothelial cells in model group were badly damaged with a large number of  inflammatory cells infiltration,and the  vascular endothelial cells in high dose of SMYA group showed integrity with  little inflammatory cells infiltration.Conclusion SMYA has a good therapeutic effect on rat model of TAO,and the mechanism may be related to anti-inflammatory and  maintaining the balance function of TXA2 and PGI2 in plasma.
