热休克蛋白融合蛋白对人树突状细胞分化成熟的刺激作用

摘要: 目的:研究热休克蛋白融合蛋白对人树突状细胞分化成熟的刺激作用。 方法:采用GM-CSF和IL-4将人外周血单核细胞诱导成未成熟树突状细胞,然后用热休克蛋白融合蛋白刺激其分化成熟,以单核细胞条件培养液(MCM)和模拟的单核细胞条件培养液(MCM mimic)为对照。以流式细胞仪检测树突状细胞表面分子的表达情况。 结果:热休克蛋白融合蛋白可以刺激树突状细胞表面表达CD40、CD80、CD86和HLA-A2分子。结论:热休克蛋白融合蛋白可诱导人树突状细胞分化成熟。

Abstract: Objective To study the effects of heat-shock protein (HSP) fusion protein on the differentiation and maturation of human monocyte derived dendritic cells (DC). Methods Human peripheral blood mononuclear cells (PBMC) were induced into immature dendritic cells (iDC) in the presences of GM-CSF and IL-4. On the 5th day the iDC was stimulated with HSP-fusion protein. Monocyte conditioned medium (MCM) and MCM mimic, a cocktail of IL-1β,IL-6,TNF-α and PGE2 were used as control. On the 7th day the DC stimulated with the HSP-fusion protein was stained with FITC-labeled antibodies against CD40, CD80, CD86, and HLA-A2 separately and the results were determined by FACS. Results HSP-fusion protein could up-regulate the iDC to express CD40, CD80, CD86, and HLA-A2 molecules. Conclusion HSP-fusion protein can induce the differentiation and maturation of human DC.
