新生小牛视网膜水平细胞生长发育特性的体外实验

摘要: 目的:研究新生小牛视网膜水平细胞在体外培养中的生长发育规律。 方法:分离培养新生小牛视网膜神经细胞,将培养10、20、30和40 d的细胞进行免疫细胞化学染色,采用Calbindin-D28K抗体标记视网膜水平细胞,用图像分析系统定量测量阳性细胞的免疫反应强度。 结果:培养10 d的水平细胞还不具备典型的形态特征,培养20 d时阳性细胞的形态特征与成熟的水平细胞一致。培养30 d和40 d的水平细胞的形态特点和培养20 d时相似。定量分析显示体外培养20 d时阳性细胞免疫反应强度较10 d时有增加趋势。体外培养30 d和40 d时,阳性的水平细胞免疫反应强度较培养10 d和20 d时均明显减弱。 结论:体外培养20 d的视网膜水平细胞具备成熟的形态特征,免疫反应强度较强,是发育良好的神经元。

Abstract: Objective To investigate the developmental changes of neonatal bovine retinal horizontal cells in vitro. Methods Dissociated neonatal bovine retinal cells were cultured. Cells grown for 10, 20, 30 and 40 days in vitro were used for immunocytochemical research. Retinal horizontal cells were identified by Calbindin-D28K antibody and the immunoreactivity of stained cells was measured by image analysis system. Results The retinal horizontal cells grown for 20 days in vitro had mature morphological characteristics. The morphology of neurons grown for 30 days and 40 days in vitro was same as that of 20 days in vitro. The immunoreactivity of the stained cells cultured for 20 days in vitro trended to increase when compared with that of 10 days in vitro. The immunoreactivity of cells cultured for 30 days and 40 days in vitro decreased significantly when compared with that of 10 days and 20 days in vitro resp ectively. Conclusion The horizontal cells cultured for 20 days in vitro have mature morphological characteristics and stronger immunoreactivity. They are well developed neurons in vitro.
