海人酸杏仁核点燃癫痫大鼠海马和顶叶皮层脑电图改变

摘要: 目的:记录和比较海人酸(KA)杏仁核点燃癫痫大鼠海马和顶叶皮层脑电图。方法:选用雄性Wistar大鼠,以右侧杏仁核外侧基底核为给药靶点,在立体定位仪下,微量注射KA建立点燃癫痫模型,在对大鼠行为学观察的同时,连续记录大鼠点燃后6 h左侧海马 和顶叶皮层脑电图。结果:大鼠点燃后脑电图依次出现多种形式的癫痫放电,以多发性棘波为主。海马和顶叶癫痫放电潜伏期,每小时放电持续时间、6 h总放电时间和波幅差异均无显著性。结论:KA杏仁核点燃大鼠脑电图改变符合颞叶癫痫特征,大鼠点燃急性早期癫痫放电在皮层间传播迅速,点燃灶对侧海马和顶叶皮层脑电图具有高度一致性,监测顶叶皮层脑电图可能更适用于对该模型的电生理学观察。

Abstract: Objective To record the electroenciphalogram(EEG) in hippocampus or parietal cortex and then compare them in epileptic rats kindled by kainic acid(KA) microinjection into nucleus amygdalae. Methods KA was microinjected(1 μg) into right nucleus amygdalae of male Wistar rats with stereotaxic technique to establish kindling epileptic model. Observation of praxiology was carried while recordin g of the EEG in hippocampus and parietal cortex were performed successively for 6 h after KA injection. Results Many kinds of epileptic discharges presented in the kin dled rats, especially the multiple spike waves were common. There were no significant differences in the latency to onset, period of discharge(PD) per hour, total PD in 6 h and the amplitude of wave between EEG of hippocampus and that of parietal cortex. Conclusion The changes of EEG in rats kindled by KA microinjection into nucleus amygdalae are consistent with that of temporal lobe epilepsy. Seizures are quickly spread inter-cortex. The epileptic discharges of hippoc ampus opposite to kindling focus are highly coincident to that of parietal cort ex. Motoring the EEG of parietal cortex may refer to observe the changes in electrophysiology for this model.
