摘要: 目的:研究海洛因对雌性大鼠垂体催乳素(PRL)基因表达的影响。 方法:50只雌性Wistar大鼠,随机分为5组(每组10只):对照组、海洛因给药3和9 d组、海洛因给药 9 d后停药3和9 d组;逆转录聚合酶链反应(RT-PCR)检测垂体PRL mRNA相对含量,放射免疫分析法(RIA)检测血浆催乳素(PRL)水平。 结果:给药3和9 d组及停药3 d组大鼠垂体PRL mRNA水平显著低于对照组(P<0.01),停药9 d组PRL mRNA的水平虽低于对照组,但差异无显著性(P>0.05);与给药9 d组相比,停药3 d组PRL mRNA水平有增高趋势,但差异无显著性(P>0.05),停药9 d组的PRL mRNA的水平则显著高于给药9 d组(P<0.05)。 给药3和9 d组及停药3和9 d组血浆PRL显著低于对照组(P<0.01);与给药9 d组比较,停药3 d组的血浆PRL水平仍然较低(P<0.05),停药9 d组的PRL水平有高于给药9 d组的趋势,但差异无 显著性(P>0.05)。 结论:海洛因使雌性大鼠垂体PRL mRNA表达水平下降,短期停用海洛因其无法恢复到正常水平。

Abstract: Objective To investigate effects of heroin on prolactin (PRL) mRNA in female rats. Methods Fifty female Wistar rats were randomly divided into five groups (10 rats each group): control, heroin-treated-3 d, heroin-treated-9 d, heroin-withdrawal-3 d and heroin-withdrawal-9 d groups. PRL mRNA of pituitary was assessed by RT-PCR and PRL concentration in rat plasma was determined by radioimmunoassay (RIA). Results The expression levels of PRL mRNA of pituitary were declined significantly in heroin-treated-3 d, 9 d and heroin-withdrawal-3 d groups compared with control group(P<0.01). Compared with heroin-treated- 9 d group, PRL mRNA level in heroin-withdrawal-9 d group was increased significantly(P<0.05). Compared with control group, the PRL level was lower sign ificantly in heroin-treated-3 d and 9 d groups(P<0.01);Also, the PRL levels were decreased significantly in heroin-withdrawal-3 d and 9 d groups(P<0.01). The PRL level was decreased remarkably in heroin-withdrawal-3 d group compared with heroin-treated-9 d group (P<0.05);in heroin-withdrawal-9 d group, the level of PRL was higher than that in heroin-treated-9 d group, but the difference was not significant(P>0.05). Conclusion The PRL mRNA in the pituitary and the PRL concentration in plasma in female rats are decreased by heroin and the effect can not be reversed in a short term after heroin withdrawal.
