人参皂苷Rg3对HSE模型鼠的治疗作用及免疫调节效应

Therapeutic effect of ginsenoside Rg3 on HSE mice and its immunoregulative activity

摘要: 目的:研究人参皂苷Rg3对单纯疱疹病毒性脑炎(HSE)模型鼠的治疗作用及其免疫调节效应。方法:腹腔注射单纯疱疹病毒1(HSV-1)病毒悬液,用PBS做六个稀释度10倍递次稀释,分别为10-1、10-2、10-3、10-4、10-5和10-6,制备小鼠HSE模型。硝酸还原酶法检测小鼠脑组织NO含量。腹腔注射人参皂苷Rg3,剂量分别为0.3、1.0和3.0 mg•kg-1,观察Rg3对HSE模型鼠存活时间的影响和脑组织病理形态的改变。MTT法检测Rg3对小鼠淋巴细胞转化功能和NK细胞毒活性的影响,鼠脾T淋巴母细胞增殖分析法测定上清中IL-2活性,细胞病变抑制法测定IFN-γ活性。结果:成功建立HSE小鼠模型。HSE模型鼠脑组织中NO含量高于对照组(P<0.05)。0.3 mg•kg-1Rg3组和疱疹性脑炎小鼠的存活时间延长,脑组织病理改变减轻。0.4 mg•kg-1Rg3组SI、IL-2和IFN-γ高于对照组(P<0.05),NK细胞毒活性高于对照,但差异无显著性(P<0.05)。结论:Rg3对HSE小鼠具有保护作用,具有免疫调节效应,是一种具有潜在开发前景的抗HSV-1药物。

Abstract: To investigate the therapeutic effect of ginsenoside Rg3 on herpes simplex encephalitis (HSE) mice and its immunoregulative activity. Methods HSV-1 suspension was diluted into 10-1,10-2,10-3,10-4,10-5 and 10-6 with PBS and injected intraperitoneally to establish HSE mouse model. NO in the brain homogenate was examined by nitrate reductase method. Rg3 with doses of 0.3, 1.0 and 3.0 mg•kg-1 were administered intraperitoneally and the influence of Rg3 on the mouse survival time and pathological changes of the brain tissue were observed. MTT colorimetry was used to detect the influence of Rg3 on lymphocyte transformation function and NK cytotoxicity. The activity of IL-2 was detected by lymphoblast proliferation assay and IFN-γ by cytopathic effect inhibition. Results HSE mouse model was successfully established. The content of NO in the brain homogenate was higher in the infected group than that in the control group (P<0.05). 0.3 mg•kg-1 Rg3 could extend survival time and ameliorate pathological changes in HSE mice. 0.4 mg•kg-1 Rg3 could significantly enhance the lymphocyte transformation function and improve the secretion of IL-2 and IFN-γ (P<0.05). NK cytotoxicity was also strengthened. Conclusion Rg3 has protective effect on HSE mice. It also has immunoregulative activity. Rg3 is a potent antiviral drug which deserves further development.
