PLA2G4B基因多态性与精神分裂症的关联性分析

Analysis on |association between PLA2G4B gene polymorphisms and schizophrenia

摘要:

目的:探讨中国北方汉族人群PLA2G4B基因多态性与精神分裂症的遗传关联性。方法:采用聚合酶链反应(PCR)和连接酶检测反应(LDR)方法,在201个精神分裂症患者核心家系中检测PLA2G4B基因上的错义突变位点rs3816533的单核甘酸多态性,应用遗传统计学方法对研究对象进行单倍型相对风险分析(HRR)和传递不平衡分析(TDT)。 结果:PLA2G4B基因rs3816533位点在精神分裂症患者组和父母组中基因型频数分布均符合Hardy-Weinberg平衡定律(P>0.05);HRR分析结果显示:父母传递和未传递给患病子女的等位基因频数分布差异无显著性(χ2 = 0.840,P>0.05);TDT分析结果显示:杂合子父母双亲的2个不同等位基因传递概率没有偏离50% (χ2 = 0.818,P>0.05)。 结论:在中国北方汉族人群中PLA2G4B基因rs3816533位点单核甘酸多态性与精神分裂症可能无关联。

Abstract:

Abstract:Objective To investigate the genetic association between the polymorphisms of PLA2G4B gene and schizophrenia in the North Han Chinese population. Methods The method of PCR-based ligase detection reaction (PCR-LDR) was applied to detect the genotype single nucleotide polymorphisms of rs3816533 in PLA2G4B gene among 201 pedigrees consisting of fathers,mothers and affected offsprings with schizophrenia. Haplotype relative risk (HRR) test and transmission disequilibrium test (TDT) were conducted to analyze the genotyping data. Results The genotypic frequency of rs3816533 of PLA2G4B gene did not deviate from Hardy-Weinberg equilibrium in both case and control groups. HRR analysis did not show allelic association with the PLA2G4B gene (χ2 = 0.840,P>0.05);TDT analysis did not show preferential transmission of the two alleles(χ2 = 0.818,P>0.05).  Conclusion  There is no genetic association between the polymorphism of rs3816533 site on  PLA2G4B gene  and schizophrenia in Han population from the north of China.
