小剂量地塞米松对颞叶癫痫大鼠空间记忆能力的保护作用

摘要:

目的:探讨小剂量地塞米松对颞叶癫痫雄性大鼠空间记忆能力的影响,为糖皮质激素相关药物在颞叶难治性癫痫治疗中的应用奠定理论基础。方法:雄性Wistar大鼠随机分为假手术组(Sham组)、生理盐水预处理后颞叶癫痫点燃对照组(NS-对照组)和小剂量地塞米松短期预处理后颞叶癫痫点燃组(Dex组),采用海人酸杏仁核微量注射方法建立颞叶癫痫点燃模型,分别于点燃或假手术后第5、11、17和21天测定大鼠Morris水迷宫空间记忆能力。结果:NS-对照组和Dex组大鼠较Sham组平均逃避潜伏期延长(P<0.05),泳动的初始角度增大(P<0.05);Dex组大鼠较NS-对照组Morris水迷宫泳动中心区域增大(P<0.05),泳动路径缩短(P<0.05)。结论:小剂量地塞米松对颞叶癫痫大鼠空间记忆能力具有保护作用。

Abstract:

Abstract:Objective
To investigate the influence of low-dose dexmethasone (Dex)on spatial memory in temporal lobe epileptic male rats,and lay theoretical  foundation for the application of glucocorticoid related medicine in refractory temporal lobe epilepsy. Methods Wistar rats were randomly divided into Sham group,control group kindled with temporal lobe epilepsy after pretreated with normal saline (NS-control group) and observation group kindled with temporal lobe epilepsy pretreated with low-dose Dex (Dex group). The temporal lobe epileptic model was established through  microinjecting kainic acid (KA) into right nucleus amygdalae of Wistar male rats. Morris water maze observation was performed on the 5th,11st,17th,and 21st days after KA kindling or sham operation,respectively. Results The average escape latency in Sham group was delayed compared with  Dex group or NS-control group (P<0.05),the swimming initial angles in Dex group or NS-control group were enlarged compared with  Sham group (P<0.05). Compared with NS-control group and Sham group,the percentage of swimming center in Dex group was increased(P<0.05)and the length of swimming path was decreased(P<0.05). Conclusion Low-dose Dex may have protective effect on spatial memory in temporal lobe epileptic male rats.
