CTLA4-Ig对支气管哮喘患者外周血IL-13的影响

Effects of CTLA4-Ig on IL-13 in peripheral blood of patients with bronchial asthma

摘要: 目的:通过对支气管哮喘患者外周血T细胞表达IL-13的情况及CTLA4-Ig对其影响的研究,进一步明确支气管哮喘的免疫发病机制并为治疗支气管哮喘寻求一条安全有效的途径。 方法:用ELISA和逆转录PCR方法测定支气管哮喘患者外周血在CTLA4-Ig作用前后T细胞表达IL-13的情况。 结果:ELISA法测得哮喘患者IL-13明显高于正常对照组(P<0.01),CTLA4-Ig明显抑制了IL-13的分泌(P<0.01)。 正常对照组PHA刺激条件下用逆转录PCR方法测不出IL-13的表达;哮喘组在PHA作用下,T细胞表达IL-13的量增加(P<0.01);同在PHA刺激情况下,CTLA4-Ig使IL-13表达明显下降(P<0.01)。 结论:IL-13在支气管哮喘发病中起重要作用,CTLA4-Ig对T细胞产生IL-13起抑制作用,对哮喘的治疗具有临床意义。

Abstract: Objective To study the expression of IL-13 in T cell from peripheral blood of bronchial asthma patients and the effects of CTLA4-Ig on it in order to study further the pathogenesis of bronchial asthma and to search for a safty and effective route to treat bronchial asthma. Methods The methods of ELISA and RT-PCR were used to determine the expressions of IL-13 in T cell from periferal blood of asthma patients before and after CTLA4-Ig treatment. Results The expression of IL-13 in bronchial asthma patients was significantly higher than normal control group (P<0.01). CALA4-Ig inhibited remarkably the secretion of IL-13 (P<0.01). The expression of IL-13 couldn′t be obtained in normal control group with PHA stimulation, and it was increased in asthma group (P<0.01), and CTLA4-Ig could decrese the exprension of IL-13 obviously after PHA stimulation (P<0.01). Conclusion IL-13 can play an important role in the pathogenesis of bronchial asthma, CTLA4-Ig can inhilit the express of IL-13 in T cell and there is clinical significance for asthma treatment.
