摘要: 目的:研究黏附分子在外周血干细胞动员机制中的作用。 方法:应用流式细胞仪检测粒细胞集落刺激因子G-CSF动员的外周血细胞与骨髓CD34+细胞表达的黏附分子非常延迟抗原4(VLA-4)、淋巴细胞功能相关抗原1(LFA-1)和L-选择素(CD62L)。 结果:G-CSF动员后的外周血CD34+细胞表达的VLA-4、LFA-1与骨髓CD34+细胞相比明显减少,CD62L无明显改变。 结论:黏附分子VLA-4及LFA-1降低可能是G-CSF动员造血干细胞进入外周血过程中的一个重要环节。

Abstract: Objective To study the value of adhesion molecules in the mechanism of peripheral blood stem cell mobilization. Methods Flow cytometry was used to detect the expressions of adhesion molecules of very late antigen 4 (VLA-4),leukocyte function-associated antigen 1 (LFA-1) and L-selectin (CD62L) on CD34+cells from granulocyte colony-stimulating factor (G-CSF)-mobilized peripheral blood(PB) and bone marrow(BM). Results The expressions of VLA-4 and LFA-1 were decreased significantly on CD34+cells from G-CSF-mobilized peripheral blood than those from bone marrow, and the expression of CD62L showed no significant difference. Conclusion The reduction of adhesion molecules of VLA-4 and LFA-1 may be an important step during the course of G-CSF-mobilized hematopoietic stem cell into peripheral blood.
