蛋白质定量中干扰因素的配伍组方差分析

摘要: 目的:探讨总体分析分离菌体蛋白的部分常用试剂对染料结合法和二辛可宁酸法蛋白质定量方法的总体干扰作用。方法:配伍组方差分析法。 结果:含EDTA和葡萄糖的分离试剂对二辛可宁酸蛋白微量定量方法干扰较强(P<0.05),盐酸胍和尿素无干扰(P>0.05);这些试剂对染料结合法蛋白微量定量方法无干扰(P>0.05)。 结论:试剂对蛋白质定量的干扰作用不应忽视。

Abstract: Objective To analyze the effects of some routine reagents of coliform protein on two kinds of protein qualification (dyebinding method and bicinchoninic acid method) was used.Methods Two way analysis of variance (two way ANOVA) was used. Results Bicinchoninic acid method was interfered by the reagents containing EDTA or glucose(P<0.05). No interference to the method was shown by using the reagents containing guanidine-HCl or urea. As for dyebinding method, all above reagents had not interference effects. Conclusion The interference of reagents can not be neglected in protein qualification.
