Effects of advanced glycation end products on peroxisome proliferator-activated receptor-γ mRNA expression in rat renal cortex

摘要: 目的:观察糖基化终末产物(AGEs)对大鼠肾皮质过氧物酶体增殖体激活受体-γ(PPAR-γ)mRNA表达的影响。方法:雄性Wistar大鼠20只随机分为4组(每组5只),AGEs组 (给予AGEs同时尾静脉注射AGE-修饰大鼠血清蛋白),AGEs+AG组[给予AGEs同时腹腔注射氨基胍(AG)],天然血清组(注射天然大鼠血清蛋白)和空白对照组(不施加处理的正常大鼠)。各组大鼠每日注射1次,连续注射6周,RT-PCR 检测PPAR-γ mRNA 表达水平。结果:各组大鼠肾皮质均有PPAR-γ表达;与天然血清组及空白对照组比较,AGEs组大鼠肾皮质PPAR-γ mRNA表达明显降低 (P<0.01), 而同时注射AG的大鼠PPAR-γ mRNA表达水平无明显降低。结论:大鼠肾皮质表达PPAR-γ;AGEs能降低大鼠肾皮质PPAR-γmRNA的表达水平,提示AGEs可能通过对肾脏保护性因子PPAR-γ的影响参与糖尿病肾病的发病机制。

Abstract: To investigate the effects of advanced glycation end products (AGEs) on expression of peroxisome proliferator-activated receptor-γ(PPAR-γ) mRNA in rat renal cortex. Methods Normal rats were given tail vein injection with either AGE-modified rat serum protein(AGEs)or AGE-RSP followed by intraperitoneal injection of AG(AGEs+AG)or native rat serum protein(native RSP). Normal rats without any treatment were as controls (Control). PPAR-γ mRNA expression was analysed by reverse transcriptase-polymerase chain reaction (RT-PCR). Results PPAR-γ mRNA expressed in all rat kidney cortex. There was a decrease for PPAR-γ in mRNA levels in the renal cortex of AGEs- treated rats(P<0.01). AG treatment prevented partly mRNA changes. ConclusionPPAR-γ mRNA signaling pathways are in rat kidney cortex. AGEs reduce the expression of PPAR-γ in renal cortex and AG inhibits the effect of AGEs. These results support the notion that modulation of PPAR-γ by AGEs may be relevant to the development of diabetic nephropathy (DN).
