解毒通络保肾胶囊对2型糖尿病大鼠血清脂联素、肾组织脂联素及MCP-1蛋白表达水平的影响

Effects of JieDu TongLuo BaoShen capsule on serum adiponectin and expression levels of  adiponectin and MCP-1 proteins in kidney tissue of type 2 diabetic rats

摘要:

[摘 要] 目的:探讨解毒通络保肾胶囊(保肾胶囊)对2型糖尿病大鼠肾组织脂联素及单核细胞趋化蛋白1(MCP-1)蛋白表达的干预作用,阐明其肾脏保护作用及机制。方法:将65只雄性Wistar大鼠随机分为正常对照组(对照组,10只,喂饲常规大鼠饲料)、高脂膳食组(高脂组,10只)和糖尿病组(45只),后2组大鼠实验期间喂饲高脂饲料。4周后糖尿病组大鼠腹腔注射低剂量链脲佐菌素复制2型糖尿病模型,造模后再随机分为糖尿病模型组(模型组)、保肾胶囊组(1.4 g/kg)和罗格列酮联合贝那普利组(阳性对照组)。连续进行相应处理12周后,测定大鼠血糖(BG)、血清肌酐(Scr)、尿素氮(BUN)、尿微量清蛋白排泄量(UAE)及血清脂联素水平,免疫组织化学法检测肾组织脂联素和MCP-1蛋白表达水平。结果:与对照组比较,保肾胶囊组糖尿病大鼠BG、Scr、BUN水平及UAE明显降低(P<0.01)。与对照组比较,模型组大鼠血清脂联素水平、肾组织脂联素蛋白表达水平明显降低(P<0.01),肾组织MCP-1蛋白表达水平明显升高(P<0.01)。保肾胶囊组大鼠血清脂联素水平和肾脂联素蛋白表达水平较模型组分别升高1.5和3.0倍(P<0.01),而肾组织MCP-1蛋白表达水平降低55%(P<0.01)。相关分析,血清脂联素水平与UAE(r=-0.466,P<0.01)、Scr水平(r=-0.563,P<0.01)、BUN水平(r=-0.512,P<0.01)及肾组织MCP-1蛋白表达水平(r=-0.526,P<0.01) 均呈负相关关系。结论:保肾胶囊处理可升高2型糖尿病大鼠血清脂联素水平,上调肾组织脂联素水平和下调肾组织MCP-1蛋白表达水平,这可能与保肾胶囊的肾脏保护作用有关。

Abstract:

Abstract:Objective  To investigate the intervene effect of JieDu TongLuo BaoShen capsule (BaoShen capsule) on the  expressions of  adiponectin protein and monocyte chemoattractant protein-1 (MCP-1) in kidney tissue of  type 2 diabetic rats,and to clarify the renoprotective effect of BaoShen capsule and its mechanism.Methods Sixty-five male Wistar rats were divided randomly into normal control group (control,10 rats,fed standard rat chow),high-fat diet feeding group(High-fat control,10 rats,fed high-fat diet) and diabetic group(45 rats,fed high-fat diet).Four weeks later,type 2 diabetes was induced in the  rats in diabetic group by intraperitoneal injection of a low dose of streptozotocin.After modeling they were again randomly divided into untreated diabetic model group(DM,15 rats),BaoShen capsule(1.4 g/kg) treatment  group (BaoShen capsule,15 rats) and rosiglitazone combined with benazepril treatment  group (Positive control,15 rats),and treated for 12 successive weeks while continuing on the high-fat diet.Twelve weeks later,the blood glucose (BG),serum creatinine (Scr) and blood urea nitrogen(BUN) levels,urinary microalbumin excretion rate (UAE) were measured.The protein expression of  adiponectin in kidney tissue was determined using immunohistochemistry.Results Compared with control group,the  blood glucose,serum Scr and BUN levels,and UAE of the  rats in BaoShen capsule treatment group were significantly decreased(P<0.01);the serum adiponectin level  and the protein expression of  adiponectin in kidney tissue of the rats in DM group were markedly reduced   compared with control group(P<0.01),and the protein expression of  MCP-1 in kidney tissue was obviously elevated (P<0.01). Compared with DM group,the serum adiponectin level  and protein expression level of  adiponectin in kidney tissue in Baoshen capsule group were increased by 1.5 and 3 folds (P<0.01),while the MCP-1 protein expression level in kidney tissue was decreased by 55% (P<0.01).Correlation analysis indicated that the  serum adiponectin levels were negatively correlated with UAE(r=-0.466,P<0.01),Scr (r=-0.563,P<0.01),BUN (r=-0.512,P<0.01) levels and  MCP-1 protein expression level in kidney tissue (r=0.526,P<0.01).Conclusion BaoShen capsule treatment can increase the serum adiponectin level,up-regulates the adiponectin level in kidney tissue and and down-regulate the MCP-1 protein expression level in kidney tissue in type 2 diabetic rats,that is linked to renoprotective capacity of BaoShen capsule.
