嘌呤对慢性吗啡处理大鼠热痛阈与急性戒断的影响

Effect of purine on thermo-pain threshold and acute withdrawal in chronic morphine treated rats

摘要: 目的:研究嘌呤碱基与嘌呤核苷对吗啡依赖大鼠的热痛阈与急性戒断的影响。 方法:28只大鼠随机分成对照组、吗啡依赖组、吗啡依赖+嘌呤碱基组、吗啡依赖+嘌呤核苷组,每组7只。通过热敏感甩尾实验,记录大鼠热敏感甩尾潜伏期;通过纳洛酮催促,对大鼠急性戒断症状进行观察与评定。 结果:与对照组相比,吗啡依赖组、吗啡依赖+嘌呤碱基组、吗啡依赖+嘌呤核苷组均产生明显的药物抗痛觉效应(P<0.05);吗啡与嘌呤碱基或与嘌呤腺苷同时作用大鼠的甩尾潜伏期与吗啡依赖组相比有延长的趋势(P>0.05);吗啡与嘌呤碱基或与嘌呤核苷同 时作用的大鼠戒断后,湿狗样抖动及腹泻/排便的发生次数比吗啡单独作用组明显增加(P<0.05)。 结论:本实验观察到系统应用腺嘌呤/鸟嘌呤、腺苷/鸟苷对吗啡依赖大鼠既可提高其基础热痛阈,又可加重其部分急性戒断症状。

Abstract: Objective To determine the effect of purine bases and purine nucleosides on thermo-pain threshold and acute withdrawal in morphine dependent rats. Methods Twenty-eight Wistar rats were randomly divided into 4 groups: control group, morphine dependent group, morphine plus purine bases group and morphine plus purine nucleosides group, there were 7 rats per group. The tail flick test was used to determine the thermo-analgesic latency and naloxone was used to observe and evaluate the precipitated syndrome. Results In morphine, morphine plus adenine and guanine, and morphine plus adenosine and guanosine groups, significant antinociceptive effects were observed as compared with control group(P<0.05). In morphine plus aden ine and guanine, and morphine plus adenosine and guanosine groups, the latencies of tail flick were increased, while the counts of wet dog shakes and diarrhea or defecation were significantly higher than those in morphine dependent rats(P<0.05). Conclusion Adenine and guanine, as well as adenosine and guanosine can raise the basic thermo-analgesic threshold in chronic morphine dependent rats. Co-adminstration of large dosage of adenine and guanine or adenosine and guanosine together with morphine may aggravate some of the withdrawal symptoms.
