西比灵对神经元损伤的保护作用

Protective effects of Sibelium on neuron injury in rats

摘要: 目的:探讨西比灵对神经元损伤的保护作用机制。 方法:采用H2O2建立离体培养大鼠皮层神经细胞损伤模型,利用八木法(TBA法)测定丙二醛(MDA)、二苯胺法测DNA断裂率,同时观察乳酸脱氢酶(LDH)泄漏率的变化、西比灵对上述指标的影响及其在体外与自由基的相互作用。 结果:与H2O2损伤组比较,西比灵可明显降低H2O2对神经元的过氧化损伤作用,LDH泄漏率、DNA断裂率明显降低(P<0.01,P<0.05),MDA含量明显下降(P<0.01),而且随着西比灵浓度的提高羟自由基清除率明显升高。 结论:西比灵不但可防止Ca2+过量跨膜进入细胞内,而且对神经元过氧化损伤具有明显保护作用。

Abstract: Objective To study the protective effects of Sibelium on neuron injury in rats and its mechanism. Methods The damage model of cultured cortical neuron induced by H2O2 was established. The malondialdehyde (MDA) was detected by TBA method, and the breaking rate of DNA was determined by diphenylamine method. The change of transudatory rate of lactate dehydrogenase (LDH), the effects of Sibelium on those indexes described above and its interaction with •OH in vitro were observed. Results The intoxication of H2O2 was decreased by Sibelium. Sibelium singnificantly reduced the transudatory rate of LDH, the breaking rate of DNA. The content of MDA in culture and the clearance rate of •OH was increased with the increasing of the dosage of Sibelium. Conclusion Sibelium can obviously protect the cultured cortical neuron against intoxincation of H2O2 in rats.
