荞麦黄酮复方制剂对糖尿病大鼠肾脏组织形态学变化的抑制作用

Inhibitory effect of flavones of buckwheat compound on histomorphologic changes of kidney tissue in diabetic rats

摘要:

目的:采用光镜及电镜技术观察荞麦黄酮复方制剂(FBC)对糖尿病大鼠肾组织形态学变化的抑制作用,初步探讨FBC防治糖尿病肾病(DN)的作用机制。方法:用高脂饲料和小剂量链脲佐菌素(STZ)建立雄性SD大鼠2型糖尿病模型,70只大鼠随机分为模型对照组(n=13,蒸馏水10 mL/kg/d),阳性药物组(n=12,消渴丸0.83 g/kg/d),FBC低、高剂量组(n=13,FBC 0.6、1.2 g/kg/d)和正常对照组(n=12,蒸馏水10 mL/kg/d),各组大鼠灌胃给药连续8周。实验结束时,用血糖仪测空腹血糖(FBG)、双肾指数,通过HE和Masson染色光镜下观察肾脏组织形态,电镜下观察肾组织超微结构。结果:与模型组比较,FBC 低、高剂量组大鼠FBG和双肾指数均明显下降(P<0.01)。与正常对照组比较,FBC 低、高剂量组大鼠各指标未出现明显改变;光镜及电镜观察结果显示,FBC能够抑制糖尿病大鼠系膜基质增加和基底膜增厚,减轻肾小球硬化程度和肾小管上皮细胞空泡变性。结论:FBC对糖尿病大鼠肾组织病理和超微结构的改变有明显的抑制作用,其机制可能是与其抑制系膜细胞增生及肾小球基底膜的生成、改善糖尿病性肾小球体积增大及硬化有关。

Abstract:

Abstract:Objective To evaluate the inhibitory effect of Flavones of Buckwheat Compound(FBC )on the histomorphologic changes of the kidney tissue in type 2 diabeticrats,and to explore the mechanism of FBC on diabetic nephropathy(DN).Methods The type 2 diabetic male SD rat model was induced by feeding high-fat diet and intraperitoneal injection with streptozotocin (STZ).The rats were randomly divided into five groups: model group(distilled water,10 mL/kg/d),positive medicine group(XKW,0.83 g/kg/d),low and high doses FBC groups(FBC,0.6 and 1.2 g/kg/d),and normal control group(distilled water, 10  mL/kg/d).The rats in each group  were gavaged daily for eight weeks.At the end of the experiment,the level of fasting blood glucose(FBG) was detected and the index of kidney was observed.HE and Masson staining were used to observe the morphological changes of kidney tissue under optical microscope.The changes of the kidney tissue ultrastucture were observed using   electron microscope.Results  Compared with model group,the level of FBG and kidney index in FBC groups were significantly decreased(P<0.01).However there were no obvious differences in these indexes between control group and FBC groups.And FBC could inhibit the increasing of mesangial matrix and the thickening of basement membrane in kidney of diabetic rats,and relieve glomerular sclerosis and renal tubular epithelial cell vacuolation.Conclusion FBC could inhibit the increasing of mesangial matrix and basement membrane thickening of diabetic rats.The mechanism is related to inhibiting mesangial cell proliferation and glomerular basement membrane formation and improving the increasing of diabetic glomerular volume and hardening.
