降脂合剂对高脂血症大鼠血脂及血流异常的调节和抑制作用

Regulation and inhibition of lipid-lowering mixture on blood lipid and blood flow abnormalities of hyperlipemia rats

摘要:

目的:探讨降脂合剂对高脂血症大鼠血脂谱及血液流变学的影响,阐明降脂合剂调节血脂及血流动力学的作用机制。方法:将雄性Wistar大鼠60只随机分为正常对照组、模型对照组、辛伐他汀组和降脂合剂高、中、低剂量组,每组10只。除正常对照组用基础饲料喂养外,其余各组大鼠喂饲高脂饲料建立高脂血症大鼠模型,造模同时预防给药,空白对照组及模型对照组大鼠给予生理盐水2 mL灌胃,辛伐他汀组大鼠给予7.2×10-4g/mL的辛伐他汀混悬溶液2 mL灌胃,降脂合剂低、中、高剂量组大鼠分别给予0.25、0.50、1.00 g/mL的中药煎剂2 mL灌胃,以上给药均为每日1次,持续喂养及给药10周。10周后6组大鼠均摘眼球取血,全自动生化分析仪测定大鼠血脂谱,全自动血液流变分析仪测定大鼠血液流变学的变化。结果:与模型组比较,降脂合剂各剂量组大鼠预防用药后,胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)和载脂蛋白B(ApoB)的水平均不同程度降低,高密度脂蛋白(HDL-C)、载脂蛋白A(ApoA)的水平及apoA/apoB比值均有不同程度的升高,差异均有统计学意义(P<0.05或P<0.01);与模型组比较,降脂合剂各组大鼠血浆黏度(ηp)、全血黏度(ηb)(低、中、高切)、红细胞聚集指数(IEA)、红细胞压积(HCT)及红细胞电泳时间(EET)降低,红细胞变形指数(IED)升高,差异均有统计学意义(P<0.05或P<0.01)。结论:降脂合剂能够调节实验性高脂血症大鼠血脂代谢及血液的黏、聚、凝状态,对血脂和血流异常有较好的调节和抑制作用。

Abstract:

Abstract:Objective  To study the effect of Lipid-lowering Mixture(LM) on blood lipid profile and blood hemorheology of hyperlipidemia rats,and to elucidate the mechanism of LM on regulating blood lipid and hemorhelogy. Methods  60 male Wistar rats were randomly divided into  normal control group, model control group,simvastatin group and LM high,middle, low doses groups,and there were 10 rats in each group.The rats in normal control group were fed with normal  diet,and  the rats in the other groups were fed with a high fat diet to establish hyperlipidemia rat models, and during the modeling the drugs were  used for prevention.2 mL physiological saline were given to the rats in control group and model group,and the rats in simvastatin group  were given 7.2×10-4g/mLsimvastatin suspension solution 2 mL, and the rats in LM low, middle,high doses groups were given 0.25,0.50,1.00 g/mL decoction 2 mL,the once  daily dose  lasted for 10 weeks.After 10 weeks,the rats in 6 groups were  eyeballed blood, the blood lipid profile and blood hemorheology of each  animal in various groups were measured by full automatic biochemical analyzer and full automatic blood analyzer,respectively.Results Compared with model group, after  treatment, the levels of the cholesterol (TC ), triglyceride ( TG ),low density lipoprotein ( LDL-C ) and  apolipoprotein B ( ApoB ) in LM  groups were reduced; the high density lipoprotein ( HDL-C ), apolipoprotein A ( ApoA ) levels and apoA / apoB ratio were increased, the differences were statistically significant ( P<0.05 or P<0.01).Compared with model group, the plasma viscosity(ηp), whole blood viscosity ( low, medium, high cut ) (ηb), index of erythrocyte aggregation ( IEA ), hematocrit (HCT ),and erythrocyte electrophoresis time ( EET ) in LM groups were decreased, the index of erythrocyte deformability ( IED ) was increased, the differences were statistically significant ( P<0.05 or P<0.01).Conclusion LM can modulate blood lipid metabolism and blood viscosity, poly, and prothrombotic state of the experimental hyperlipidemia rats, which has better regulation and inhibition  on blood lipid and blood flow abnormalities.
