基质金属蛋白酶含量及表达与膀胱癌侵袭转移的关系

摘要: 目的:探讨基质金属蛋白酶2(MMP-2)、基质金属蛋白酶9(MMP-9)的血清含量及组织表达与膀胱移行细胞癌侵袭转移的关系。 方法:用ELISA法检测68例膀胱移行细胞癌患者及20例正常对照组血清中MMP-2和MMP-9的含量;应用免疫组织化学SP法,检测68例膀胱移行细胞癌、10例正常膀胱黏膜组织中MMP-2和MMP-9的表达及分布情况。 结果:膀胱癌患者血清中MMP-2和MMP-9含量明显高于正常对照组(P<0.01),淋巴结转移组患者血清MMP-2及MMP-9含量高于无淋巴结转移组;膀胱癌组织中MMP-2及MMP-9表达显著高于癌旁组织及正常黏膜,随着肿瘤分级的增高MMP-2和MMP-9表达显著增强(P<0.05)。 结论:MMP-2和MMP-9血清含量及组织表达与膀胱癌侵袭转移有密切关联,可以作为判断膀胱移行细胞癌预后的客观指标。

Abstract: Objective To investigate the correlation of matrix metalloproteinase-2,-9 (MMP-2,MMP-9) in transitional cell carcinoma of bladder (TCCB) with tumor invasion and metastasis. Methods The total serum MMP-2 and MMP-9 levels were detected in 68 patients with TCCB and 20 healthy controls by ELISA; the expresions and distributions of MMP-2 and MMP-9 in 68 patients and 10 normal bladder tissue were determined by immunohistochemical method. Results The levels of serum MMP-2 and MMP-9 were significantly elevated in cancer samples compared with normal (P<0.01), significant difference was found between the patients with metastasis and without metastasis. In TCCB, the expressions of MMP-2 and MMP-9 were increased significantly compared with normal bladder epithelial. The expressions of MMP-2 and MMP-9 were correlated with histological grade and lymph node metastasis (P<0.05).  Conclusion These results suggest that the total levels of serum MMP-2 and MMP-9 and their expressions may provide reliable information in TCCB prognosis.
