含鼠GM-CSF基因的腺病毒载体对荷B16黑色素瘤小鼠免疫功能的影响

Effects of carrying murine GM-CSF gene adenovirus vectoron immune response of B16 melanoma bearing mice

摘要: 目的:研究含鼠GM-CSF基因的腺病毒载体(AdCMVGM-CSF)直接注射荷B16黑色素瘤小鼠瘤体内对小鼠免疫功能的影响。 方法:将B16黑色素瘤细胞(5×106/只)接种于C57/BL6小鼠右侧背部皮下后,将荷瘤小 鼠随机分为2组,每组12只,待瘤体直径达5 mm时分别注射AdCMVGM-CSF、AdCMVLacZ (5×108 PFU/只),检测每组小鼠的免疫功能及其对B16黑色素瘤的抑制作用。 结果:AdCMVGM-CSF实验组小鼠注射后第10天,荷瘤鼠血清中GM-CSF水平[(1 563.72±136.58)ng·L-1]高于对照组(P<0.01),肿瘤组织周围的CD8+ T细胞表达明显增强。 结论:AdCMVGM-CSF可增强荷B16黑色素瘤小鼠的抗肿瘤免疫反应,具有抑制肿瘤生长的作用。

Abstract: Objective To observe the effects of carrying murine GM-CSF gene adenovirus vector (AdCMVGM-CSF) on immune response of B16 melanoma bearing mice. Methods B16 melanoma cells were injected subcutaneously at right back of C57/BL6 mice, 24 tumor bearing mice were divided into 2 groups at random, when tumor diameter attained to 5 mm, AdCMVGM-CSF and AdCMVLacZ were injected in different groups (5×106/mouse), respectively, immune responses of each group were tested, and the inhibitory effects of AdCMVGM-CSF and A dCMVLacZ on B16 melanoma were observed. Results Compared with control group, serum GM-CSF level attained to top in experiment group at 10th day,was (1 563.72±136.58)ng·L-1 (P<0.01), the expression of CD8+ T lymphocyte around tumor tissue was high. Conclusion AdCMVGM-CSF can enhance the anti-tumor immune response of B16 melanoma bearing mice and inhibit the growth of tumor.
