骨髓增生异常综合征患者骨髓细胞增殖活性的研究

Study on proliferation of hematopoietic cellin patients with myelodysplastic syndrome

摘要: 目的:研究骨髓增生异常综合征(MDS)患者骨髓细胞增殖活性的改变,以探讨MDS疾病本质。方法:采用体外液体单层和半固体培养法及流式细胞分析术。结果:①MDS/RA的CFU-GM、CFU-E产率显著低于对照组(P<0.001),而MDS/RAEB患者未检测到CFU-GM、CFU-E集落形成;②MDS患者总体G0/G1期细胞百分比明显高于对照组(P<0.01),而S、G2+M期细胞百分数均低于对照组(P<0.01);③低危组MDS/RA凋亡与正常对照组相比较明显增高(P<0.01),而高危组MDS/RAEB凋亡与正常对照组相比较差异无显著性;MDS患者总体PCNA表达明显高于正常对照组(P<0.01),高危组MDS/RAEB PCNA表达增高尤为显著;④MDS患者P53蛋白表达与正常对照组相比较明显增高(P<0.01),BCL-2蛋白表达与正常对照组相比较明显降低(P<0.01)。结论:MDS患者的造血干细胞异常,而且MDS患者存在高增殖性和过度凋亡,p53与bcl-2基因参与了凋亡调控。

Abstract: Objective To study the proliferation of hematopoietic cell in patients with myelodysplastic syndrome (MDS). Methods Liquid single layer culture and semisolid culture in vitro were used and flow cytometry was used to detect the cell cycle, apoptosis and the expressions of PCNA, P53,and BCL-2. Results ①The Clony Forming Unit-Granulocyte-Macroplage(CFU-GM) and the Erythroid-Clony Forming Unit(CFU-E) of myelodysplasyic syndromes (MDS/RA) decreased significantly (P<0.001); while CFU-GM,CFU-E of MDS/RAEB could not be detectable; ②The percentage of G1 phase increased and the percentage of S, G2/M phase decreased significantly (P<0.01), compared with control group; ③The increase of apoptotic bodies of MDS/RA was much more markedly (P<0.01), while the changes of those in MDS/RAEB could not be found significantly; the expression of PCNA of MDS increased in both MDS/RA and MDS/RAEB (P<0.01), especially in MDS/RAEB group;④the protein expression of P53 in MDS increased, while BCL-2 decreased significantly in MDS, compared with normal control (P<0.01). Conclusion There are severe abnormalities in hematopoietic cell of patients with MDS, higher proliferation and excessive apoptosis occur in bone marrow cells and the changes of p53,bcl-2 gene suggest their possible participation in the regulation of apoptosis.
