Effects of LaCl3 on P16 and P21 expressions of hepatocellular carcinoma cells

摘要: 目的: 研究三氯化镧(LaCl3)对大鼠肝癌细胞P16和P21蛋白表达的影响。法:实验分为4组,3个实验组:CBRH-7919细胞分别培养在含0.01、0.10和1.00 mmol•L-1 LaCl3的DMEM培养液中;对照组:CBRH-7919培养在不含LaCl3的DMEM培养液中。培养时间分别为1、3和5 d。观察细胞生长变化,运用流式细胞术检测细胞周期变化,采用免疫细胞化学方法检测与G1期调控有关的P16和P21的变化情况。结果:与对照组比较,0.10和1.00 mmol•L-1LaCl3组培养后3 和 5 d CBRH-7919细胞的生长抑制率均明显增加(P< 0.01),G0/G1期细胞百分数显著增加(P< 0.01),P16和P21阳性率明显增加(P< 0.01)。 结论:LaCl3可通过上调P16和P21,使肿瘤细胞从G1期进入S期受阻,从而抑制CBRH-7919细胞生长。

Abstract: To study the effects of LaCl3 on P16 and P21 expressions of hepatocellular carcinoma cells. Methods The experiment was divided into 4 groups.In experiment groups,CBRH-7919 cells were cultivated with 0.01,0.10 and 1.00 mmol•L-1 LaCl3 in DMEM containing 10% calf serum.In control group,CBRH-7919 was cultivated in DMEM containing 10% calf serum without LaCl3.The growth of CBRH-7919 cells was observed following treated with 0.01,0.10,1.00 mmol•L-1 LaCl3 for 1,3,5 d in vitro, respectively,the changes of cell cycle were detected by flow cytometry.At the same time,P16 and P21 were detected by immunocytochemical analysis. Results Compared with control group,the inhibitory rate of growth of CBRH-7919 cells was increased evidently when treated with 0.1 and 1.00 mmol•L-1 LaCl3 for 3 and 5 d(P< 0.01),the percentage of CBRH-7919 cells ofG0/G1 was evidently increased(P< 0.01),the expressions of P16 and P21 were increased(P< 0.01). Conclusion LaCl3 can obviously inhibit the growth of CBRH-7919 cells by up-regulating P16 and P21 that can block the cell cycle progress from G1 to S.
