局灶性脑缺血及再灌注损伤IGF-Ⅰ表达的动态变化

Changes of IGF-Ⅰ expression in focal cerebral ischemical reperfusion injury model of rats

摘要: 目的:探讨胰岛素样生长因子-Ⅰ(IGF-Ⅰ)在局灶性脑缺血再灌注损伤中的表达及作用。 方法:采用线栓法制备大鼠局灶性脑缺血模型,应用免疫组化染色、原位杂交技术检测IGF-Ⅰ蛋白及mRNA表达。 结果:正常组织有极低水平的IGF-Ⅰ表达,脑缺血后表达主要位于半影区,随缺血及缺血再灌注时间延长,半影区表达逐渐增强,IGF-Ⅰ蛋白及mRNA表达在再灌注24 h为169.64±6.97、168.64±6.28,48 h为168.86±5.39、170.86±5.07,并达高峰。 结论:内源性IGF-Ⅰ表达增强,对局灶性脑缺血再灌注损伤具有保护作用。

Abstract: Objective To study the expression of insulin-like growth factor-Ⅰ (IGF-Ⅰ) in focal cerebral ischemical reperfusion injury model and its contribution. Methods The focal cerebral ischemia rat models were induced using the filament method, the expressions of IGF-Ⅰ protein and IGF-Ⅰ mRNA was dectected by the method of immunohistochemical staining and in situ hybridization technique. Results The expression of IGF-Ⅰ was very low in normal tissues, while increased mainly in penumbra after cerebral ischemia. The expressions of IGF-Ⅰ protein and IGF-Ⅰ mRNA were 169.64±6.97 and 168.64±6.28,respectively, at 24 h after reperfusion and peaked to 168.86±5.39 and 170.86±5.07 at 48 h after reperfusion. Conclusion Enhanced expression of endogenous IGF-Ⅰ has protective effects on focal cerebral ischemia reperfusion injury.
