XIAP基因在乳腺癌发生及发展过程中的表达及意义

摘要: 目的:探讨XIAP基因在乳腺癌发生和发展中的作用。 方法:采用免疫组化S-P法检测正常乳腺组织(96例)、乳腺囊性增生组织(56例)、乳腺不典型增生组织(12例)及乳腺癌组织(119例)中XIAP蛋白的表达,并分析其与乳腺癌临床病理特征的关系。结果:XIAP蛋白在正常乳腺组织、乳腺囊性增生组织、乳腺不典型增生组织及癌组织中的阳性率分别为4.2%(4/96)、5.4%(3/56)、42.7%(5/12)和72.2%(86/11 9),后两者的阳性表达率明显高于前两者(P<0.005);浸润性非特殊型乳腺癌XIAP蛋白阳性率(82.0%,73/89)明显高于浸润性特殊型乳腺癌(45.4%,10/22)和早期浸润性乳腺癌(37.5%,3/8)(P<0.05);XIAP蛋白表达在有淋巴结转移的乳腺癌中有升高趋势,但无统计学意义(P>0.05)。 结论:XIAP蛋白在乳腺癌发生、发展不同阶段呈不同程度的表达,过度表达提示预后不良。

Abstract: Objective To explore the effects of XIAP on carcinogenesis and progression of breast cancer.Methods The expressions of XIAP in the tissues of normal mammary (96 cases), cystic hyperplasia mammary (56 cases), atypical hyperplasia mammary (12 cases),and breast cancer (119 cases) were evaluated by SP immunohistochemistry. The relationships between XIAP expression and the pathologically biological features of breast cancer were assessed.Results The positive rates of XIAP in the tissues of normal mam mary, cystic hyperplasia mammary, atypical hyperplasia mammary and breast cancer were 4.2%(4/96), 5.4%(3/56),42.7%(5/12) and 72.3%(86/119), respectively. The positive rates in the last two groups were higher than those in the former two groups (P<0.05). XIAP was more expressed in the infiltrative non-special breast cancer (82.0%,73/89) than in the special and early stage infiltrative breast cancer (37.5%,3/8)(P<0.05). Expression of XIAP was increased in breast cancer with lymph node metastasis although the difference was not significant (P>0.05). Conclusion Overexpression of XIAP is common in carcinogenesis and progression of breast cancer. Altered expression of XIAP may contribute to carcinogenesis and progression of breast cancer by inhibiting apoptosis, its overexpression indicates worse prognosis.
