氯丙嗪协同紫杉醇对Hep-2细胞的增殖抑制作用

摘要: 目的:观察盐酸氯丙嗪与紫杉醇联合应用对人喉癌细胞株(Hep-2细胞)的增殖抑制作用及其对细胞周期各时相的影响。方法:对数生长期Hep-2细胞分为单纯紫杉醇组(3.0、6.0和12.0 mg•L-1)、氯丙嗪4.0 mg•L-1)加紫杉醇(12.0 mg•L-1)联合组和空白对照组(100 mL培养液),应用MTT比色法和流式细胞术检测各组Hep-2细胞的增殖抑制率,流式细胞仪分析单独及联合用药后Hep-2细胞的周期进程及凋亡率。结果:3.0、6.0和12.0 mg•L-1单纯紫杉醇组Hep-2细胞增殖抑制率分别为14.0%、23.9%和36.7%,随紫杉醇浓度增加细胞增殖抑制率升高,三组间及组间两两比较差异均具有显著性(P<0.05);联合组Hep-2细胞增殖抑制率为46.3%,明显高于相同剂量单纯紫杉醇组(P<0.05);单纯紫杉醇组随其用药浓度的增加G1期细胞减少,S期和G2期细胞增多,与空白对照组比较凋亡率升高(P<0.05)。结论:在一定浓度下,盐酸氯丙嗪能够增强紫杉醇诱导Hep-2细胞增殖抑制率及细胞周期进程各时相变化和凋亡率,氯丙嗪与紫杉醇联合应用具有明显的协同效应。

Abstract: Abstract:Objective To investigate the proliferation inhibition of chlorpromazine combined with taxol on Hep-2 cells (human laryngeal carcinoma cell line) and the effects on cell cycle progression. Methods Hep-2 cells at logarithmic growth phase were divided into taxol groups(3.0,6.0 and 12.0 mg•L-1),chlorpromazine(12.0 mg•L-1) comined with taxol (4 mg•L-1)group and control group (100 mL culture fluid).MTT and flow cytometry were used to detect the proliferation inhibition rates of Hep-2 cells in various groups.Flow cytometry was also used to analyze the cell cycle progression of Hep-2 cells and apoptotic rate after administration.Results The proliferation inhibition rates in 3.0,6.0 and 12.0 mg•L-1 taxol groups were 14.0%,23.9% and 36.7%,respectively,there were significant differences between three groups(P<0.05);the proliferation inhibition rate in combined group(46.3%) was higher than those in taxol groups(P<0.05).In taxol groups.the number of G1 phase cells decreased and the number of S and G2 phase cells increased with the increasing of concentration of taxol.Compared with control group,the apoptotic rates in taxol groups increased(P<0.05). Conclusion Within certain concentration range,chlorpromazine could enhance taxol-induced proliferation inhibition rate,cell cycle progression and apoptotic rate of Hep-2 cells,chlorphomaxine and taxol have significant cooperation effect.
