羊膜上皮细胞促进共培养神经干细胞存活及分化

Enhanced viability and neural differentiation of neural stemcells co-cultured with amniotic epithelial cells

摘要: 目的:探讨羊膜上皮细胞是否能在体外促进胚胎脑神经干细胞的存活及分化。 方法:Neurosphere法分离、克隆E12~14 d Wistar大鼠脑组织的神经干细胞,同时从羊膜中分离羊膜上皮细胞。将神经干细胞与羊膜上皮细胞在不同条件下共培养,通过免疫组织化学法对羊膜上皮细胞及神经干细胞的分化进行检测。 结果:羊膜上皮细胞表达神经干细胞及神经元、神经胶质细胞表面抗原;与羊膜上皮细胞共培养的神经干细胞克隆的分化细胞总数、分化为神经元的百分率及神经元初级突起长度明显高于对照组。 结论:羊膜上皮细胞与神经干细胞具有一定的同源性;羊膜上皮细胞能促进体外培养的神经干细胞的分化,且主要向神经元分化,并促进神经元初级突起的生长。提示羊膜上皮细胞为神经干细胞提供促进其分化及存活适宜的微环境。

Abstract: Objective Whether amniotic epithelial cells could promote the survival and differentiation of neural stem cells was explored in vitro. Methods Neural stem cells were dissociated and cloned from the cortical tissue of E12-14 d rats. Amniotic epithelial cells were also dissociated and purified simultaneously from the amnion. The neural stem cells and amniotic epithelial cells were co-cultured. The expression of marker of neural stem cells and differentiated cells (neuron and glial cell) in the neural stem cells and amniotic epithelial cells were detected by immunohistochemistry method. The morphological changes of neural stem cells were examined with light microscope. Results Compared with control group,the total differentiated cells and percentage of MAP-2 positive cells was significantly increased in co-cultured groups. The primary neurite of neuron-like cells in co-cultured groups were obviously larger than that in control group. Conclusion Amniotic epithelial cells can provide a supportive environment for the survival and neuronal differentiation of neural stem cells and suggest that their addition may be useful as a sustained source of trophic support to improve outcome of neural stem cells transplantation.
