Establishment and evaluation of  HPLC method for detection of concentration  of oleanolic acid-3-O-β-D- pyran glucuronide in Xinlüning capsule

Abstract:

Objective  To establish an high performance liquid chromatography(HPLC) method for the determination of oleanolic acid-3-O-β-D- pyran glucuronide concentration in Xinlüning capsule,and to examine the various factors that affect the determination of oleanolic acid-3-O-β-D-pyran glucuronide concentration in Xinlüning capsule.Methods HPLC method was used to detect the concentrantion of oleanolic acid-3-O-β-D-pyran glucuronide.  Chromatogyaphic assay was performed with HypersilC18(4.6 mm×250.0 mm,5.0 μm)column  and acetonitrile-1% glacial acetic acid (56∶44).The column temperature was set at 30℃ and the wavelength was 210 nm and the flow rate  was 1.00 mL?min-1.Results Under this condition,the recorery rate of  oleanolic acid-3-O-β-D- pyran glucuronide   was 97.4% with the relative standard deviation(RSD) of 1.4%.The calibration curve showed good linearity in the range of 14.80-26.64  μg(γ=0.999 9).Conclusion The detection  of  the concentration of oleanolic acid-3-O-β-D-pyran glucuronide in Xinlüning capsule by HPLC method is simple and accurate,which can be used as the quality control method of Xinglüning capsule.
