Identification of outer membrane proteins of Brucella Melitensis

Abstract:

Abstract:Objective To identify the major outer membrane proteins(OMPs) of Brucella Melitensis  with proteome analysis. Methods The OMPs were extracted from the culture of Brucella Melitensis and then the two dimension electrophoresis was used to separate the OMPs.By means of matrix-assisted laser descorption/ionization time of light mass spectrometry (MALDI-TOF-MS),the OMPs were identified.Peptide mass fingerprints were searched in the NCBInr database using the Program Mascot.Results Sixty seven protein spots were identified,in which Omp25 and Omp31 were identified as two main OMPs and other OMPs,such as Imp,Frp and GroEL were found.The functions of identified OMPs  included thymidylate kinase,protein precursor,NADH dehydrogenase subunit,cytosol aminopeptidase,etc.Conclusion Identification and analysis of Omp25 and Omp31 provides an important clue for understanding the  molecular mechanism of   Brucella Melitensis and developing the vaccine to prevent
 Brucella attack.
