Expressions of PTEN in oral leukoplakia and oral squamous |cellcarcinoma and |its inhibitory effect |on oral squamous cell

Abstract:

Abstract:Objective To explore the role of PTEN in the development of oral squamous cell carcinoma (OSCC) by investigating the effect of PTEN on the proliferation of oral squamous carcinoma cells and its expression in oral leukoplakia(OLK) and OSCC.Methods The eukaryotic expression vector pcDNA-PTEN was constructed successfully,and then was transfected into squamous carcinoma cell line Tca8113.After the stablely transfected Tca8113 cell lines were established,MTT and flow cytometry were used to detect the growth and apoptosis of Tca8113 cells,respectively.The endogenous protein expression level of PTEN was determined by immunohistochemical method in 25 cases of oral leukoplakia and 32 cases of OSCC.0Results The over-expression of PTEN gene in pcDNA-PTEN group obviously inhibited the growth and induced the apoptosis of Tca8113 cells compared with  control group (P<0.01).Moreover,compared with control group,the cells at G0/G1 phase were significantly increased (P<0.05),but decreased at S phase (P<0.05).In OSCC,the expression of PTEN demonstrated absent or less than normal tissues and the expression level of PTEN had a significantly positive correlation with the malignancy of OSCC(P<0.05).Conclusion As a tumor suppressor gene,the expression of PTEN could suppress the growth of OSCC cell line in vitro. PTEN might play an important role in the development of OSCC.
