Screening of |active ingredient of Stigmata maydis inhibitrg |xanthine oxidase  |and its |effect

Abstract:Objective
To extract the active ingredient of Stigmata maydis for inhibiting the xanthine oxidase(XOD)and provide the basis for the further development of new Chinese medicine in treatment of  gout. Methods The production of uric acid and superoxide generation were detemined,and the inhibitory rates of XOD after treated with different solvent extraction and different solvent depuration of Stigma maydis were calculated,then the data was   compared with positive control allopurinol. Results Compared with other groups,the inhibitory rate of XOD activity of active component purified by 50% ethanol was the highest (60.90%±1.27%).The IC50 of the production of uric acid  was 6.0 mg·L-1 and that of positive control allopurinol  was 1.10 mg·L-1.The  IC50 of the production of superoxide ions was 4.5 mg·L-1 ,and that of positive control allopurinol  was 1.05 mg·L-1.Conclusion The active ingredient of Stigma maydis which could inhibit XOD activity is purified by 50% ethanol.
