Effects of purine nucleotide compensation on purine nucleotide catabolism in heroin dependent rats

Abstract:

Abstract:Objective To explore the effect  of exogenous purine nucleotide on the catabolism of nucleotide in heroin dependent rats,and clarify the mechanism of purine nucleotide treatment for heroin dependent rats. Methods  Fifty  male Wistar rats were randomly divided into control group,heroin administration group,heroin plus AMP and GMP group,heroin plus AMP group and heroin plus GMP group. The contents of uric acid(UA),blood urea nitrogen (BUN) and creatinine (Cre) in plasma were measured,the activities of adenosine deaminase (ADA) and xanthine oxidase(XO) in plasma and brain cortex were measured by  test Kits.Results  The content  of UA in plasma and the activities of ADA,XO in heroin group were significantly higher than those  in control group and  three heroin plus purine nucleotides groups (P<0.05). The contents of BUN and Cre had no difference between various groups (P>0.05).The ADA and XO activities in plasma and brain cortex in heroin group were significantly increased compared with  control group and  three heroin plus purine nucleotides groups (P<0.05,P< 0.01 ).Conclusion Compensating purine nucleotides could ameliorate the enhancement of purine nucleotide catabolism.
