Expressions of T-bet,GATA3 and FoxP3 mRNA in PBMCs in patients with chronic hepatitis B virus infectionand their significances

Abstract:Objective
To detect the expressions of T-bet,GATA3 and Foxp3 mRNA in peripheral blood mononuclear cells(PBMCs) in patients with chronic hepatitis B(CHB) infection,and to  investigate  the influence in the development of chronic HBV infection.Methods Eighty-six in-patients were selected.According to the diagnostic criteria of CHB  prevention guidelines,they were divided into four groups:33 moderate CHB patients,21 serious CHB patients,16 severe hepatitis patients and 16 cirrhosis patients.10 healthy volunteers were used as controls.RT-PCR method was used to detect the expressions of T-bet,GATA3 and Foxp3 mRNA in PBMCs in patients with CHB.Results The expression levels  of T-bet in moderate CHB group,serious CHB group,severe hepatitis group,cirrhosis group and control group  were  0.597±0.094,0.724±0.071,0.860±0.060,0.528±0.059,and 0.514±0.071,respectively.The  T-bet levels of the patients in serious CHB and severe hepatitis  groups were higher than that in control group(P<0.05);the T-bet level of the patients in moderate CHB group was lower than that in serious CHB group(P<0.05).The expression of  T-bet in serious CHB patients  was lower than that in severe hepatitis group(P<0.05).The levels of GATA3 of the patients in moderate CHB group,serious CHB group,severe hepatitis group,cirrhosis group and control group were 0.966±0.037,0.973±0.021,0.725±0.028,0.759±0.037,and 0.535±0.028,respectively.The levels of GATA3 in patients groups  were  greatly higher than that in control group(P<0.05).The GATA3 expression levels in severe hepatitis group was significantly lower than those in moderate and serious CHB groups(P<0.05).The expression levels of Foxp3 in moderate CHB group,serious CHB group,severe hepatitis group,cirrhosis group and control group were 0.935±0.034,0.919±0.047,0.804±0.021,0.984±0.060,and 0.772±0.083,respectively.The Foxp3 expression level of the patients  in severe group was lower than those in the  other   patients groups(P<0.05).The T-bet/GATA3 ratios in moderate CHB group,serious CHB group,severe hepatitis group,cirrhosis group and control group were 0.618±0.100,0.744±0.072,1.187±0.082,0.697±0.078, and 0.963±0.133, respectively.The T-bet/GATA3 ratio of the patients  in severe hepatitis group was markedly higher than those in the other   patient groups(P<0.05).Conclusion The expressions of T-bet and GATA3 are contrary during the different courses of disease.The ratio of T-bet/GATA3 is negatively correlated with the expression of Foxp3 in the patients with chronic HBV infection.
