Objective To investigate the genetic association of DQA2 gene in the DQ supregion of MHCⅡregion with psychotic symptoms of schizophrenia. Methods The polymerase chain reaction (PCR) and restriction fragment length polymorphism (RELP) were applied in this research among the 190 Han family trios in the north of China. The chi-square (χ²) test, transmission disequilibrium test (TDT), and the haplotype-based relative risk (HRR) analysis were used to process the genotype data. Results The SNP rs2239800, a G to A base change, presented in the DQA2 locus showed significant associations with the delusion of observation (χ²=7.066, P=0.008) and also with the delusion of grandeur (P=0.045). And genotype of rs2239800 showed associations with genuine auditory hallucination (χ²=6.771, P=0.034), with delusion of observation (χ²=10.686, P=0.005), with delusion of grandeur(χ²=7.103, P=0.029), and with apathy （χ²=11.906, P=0.018）. Conclusion The DQA2 locus is very likely to be associated with such psychotic symptoms as genuine auditory hallucination, delusion of observation, delusion of grandeur and apathy.

Objective To investigate the genetic association between the GLRA1 locus in chromosome 5 with susceptibility to schizophrenia among 91 Chinese Han family trios consisting of fathers, mothers, and affected offsprings with schizophrenia. Methods Genomic DNA was isolated from the whole blood samples. The PCR-based restriction fragment length polymorphism (RFLP) technique was used to detect the single nucleotide polymorphism（SNPs）,rs1428157, which was a base mutation for C to A at the GLRA1 locus. Results The distribution of genotype frequencies of rs1428157 was not deviated from the Hardy-Weinberg equilibrium; Haplotype relative risk test (HRR) did not show a genetic association between rs1428157 and schizophrenia (χ²=0.293,P>0.05）; Transmission disequilibrium test (TDT) did not show significant biased transmission from parents to affected offspring（χ²=0.281，P>0.05）; There were significant correlations between allele frequencies and incoherence of thinking and delusion of being revealed. Conclusion The rs1428157 itself is not associated with schizophrenia, but it has significant correlation with incoherence of thinking and delusion of being revealed.

Objective To study the dose-effect relationship between X-rays irradiation and CD14 expression in mouse macrophages in vivo and in vitro. Methods The CD14 positive cells from mouse peritoneal macrophages were detected by flow cytometry (FCM) 16 hours after whole body irradiation (WBI) in vivo and from the cell line J774A.1 of mouse macrophages 4 hours after irradiation in vitro with X-rays from 0.05 Gy to 6.00 Gy. Results WBI by low dose X-rays significantly enhanced the CD14 expression in peritoneal macrophages while the doses of 2.0 to 4.0 Gy caused an opposite effect. The X-irradiation in vitro caused a similar dose-effect curve but the suppression of CD14 expression occurred at doses of 4.0 to 6.0 Gy. Conclusion Low dose irradiation can stimulate CD14 expression and high dose irradiation (>2.0 Gy) can suppress its expression both in vivo and in vitro.

Objective To study the expressions of HoxB genes in astrocytoma C6 cell lines. Methods C6 cells were cultured in vitro and the expressions of HoxB2,B4,B5-B9 mRNA were detected by RT-PCR in the presence of Hox gene degenerate primers and specific primers. Results In C6 cells, Hox genes expressed in different degrees. HoxB4 did not express, and HoxB2 expressed weakly. Conclusion In C6 cells no expression of HoxB4 and the weak expression of HoxB2 may be correlated with the origin of astrocytoma.

Objective To observe cell fusion from without(FFWO) induced by human herpesvirus 6(HHV-6). Methods Cells were overlaid with 10⁵　HHV-6-infected CBMCs (nearly 80% positive for HHV-6 late antigen by IFA) or infected with cell-free HHV-6 at 10^3-4TCID₅₀ for 60 minutes and the cells were stained with HE 2-3 days after infection.To determine whether HHV-6A induced FFWO or not, Vero cells (5×10⁵ per well) were overlaid with 10⁵ HHV-6-infected CBMCs (nearly 90% positive for HHV-6 late antigen by IFA) or infected with cell-free HHV-6 at 10^3-4 TCID₅₀ for 60 minutes, and cultured with medium supplemented with PFA (300 mg•L^-1), fixed 24 hours after infection;Vero or MT4 cells were infected with cell-free virus (HHV-6A or HHV-6B) for 1 hour, and cycloheximide was added to a final concentration of 100 mg•L^-1. After incubation at 37℃ for 2 hours, the cells were fixed and stained with HE. Results The syncytia formation induced by HHV-6A was observed 2 hours after infection; moreover, it was found in the presence of cycloheximide, indicating that HHV-6A induced FFWO in the target cells. Furthermore, the fusion event was dependent on the expression of the HHV-6 entry receptor, CD46, on the target cells membrane. Conclusion HHV-6A can induce FFWO in a variety of human cells through its entry receptor, CD46.

Objective To detect the acrosome reaction induced by recombinant human zona pellucida 3 (rhZP3) in vitro. Methods The sperm from fertilized healthy men were obtained in vitro. The spermatozoa were isolated by discontinuous density gradient centrifugation, and the capacitated spermatozoa suspension were divided into negative control group (group C, 200 μL spermatoza suspension plus 1 μL DMSO), positive control group (group A, 200 μL spermatozoa plus A23187 to the end concentration of 10 μmol•L^-1) and ZP3 group (200 μL spermatozoa plus ZP3 to the end concentration of 10 mg•L^-1). The acrosome reaction induced by rhZP3 was detected with coomassie brilliant blue staining. Results The percentages of acrosome reaction of spermatozoa in group C, group A, and ZP3 group were (5.00±1.70)%,(46.10±7.49)%, and (13.20±2.70)%，respectively. There were significant differences between various groups (P<0.05).Conclusion The rhZP3 can induce acrosome reaction of spermatozoa.

Objective To study the regulation and interference of T lymphocyte on antigen-specific IgE (SIgE) antibodies by B lymphocyte in bronchial asthma mouse model and clarify the role of T lymphocyte in the pathogenesis of asthma. Methods The asthma mouse model was set up at first, then T and B lymhphocytes were isolated from the mice spleen and were cultivated in different combinations. The supernates were collected and the specific anti-OVA antibody IgE in the supernate were tested with enzyme linked immunosorbent assay (ELISA). Results The level of SIgE secreted by B cells from asthma mouse was higher than that from the control mouse (P<0.05). After regulated by own T lymphocytes, the level of SIgE secreted by B cells from asthma mouse decreased significantly (P<0.05). With or without specific antigen, after regulated by T cells from asthma mouse, the level of SIgE secreted by B cells from the control mouse increased, but there was no significant difference (P>0.05). Conclusion T lymphocyte has a key role in regulating and controlling the pathogenesis of asthma. And T cell functional abnormality is one of the important mechanisms of asthma.

Objective To investigate the effects of benazepril on connective tissue growth factor(CTGF)expression in myocardial tissue of streptozotocin(STZ)-induced experimental diabetic rats and its mechanism. Methods A total of 34 Wistar rats were enrolled in the study, 10 of them were set as normal controls, others were used to set up the diabetes mellitus models by injecting STZ, and then equally divided into benazepril-treated group and untreated group. Blood glucose and Tcho levels levels were determined, myocardial AngⅡ was detected by immunoradiometry and myocardial CTGF mRNA was assayed by RT-PCR after 12 weeks. Results Blood glucose and Tcho increased significantly in untreated group, the levels of myocardial AngⅡ and CTGF mRNA expression were higher than those in normal control and benazepril-treated group (P<0.01). Conclusion Long lasted hyperglycemia can lead to the over-expression of CTGF mRNA in the myocardial tissue of diabetic rats. Benazepril may partly suppress it and decrease significantly the myocardial injury of diabetic rats by reducing the level of myocardial AngⅡ.

Objective To detect collagen Ⅱ in constructed articular cartilage by three-dimensional culturing system using collagen and human fibrin as supporter. Methods Chondrocytes were isolated from articular cartilage of 2-week-old rabbit, digested by collagenase Ⅱ, and mixed with bovine collagen I, human blood freeze-dry fibrinogen and thrombosin according to a certain proportion,and were cultured in vitro. Biopsies was analyzed by Masson stain, hybridization in situ using oligonucleotied probe of collagen Ⅱ and immunohistochemistry test of collagen Ⅱ after 3 weeks. Results After 3 weeks, all cells were viable, formed cartilage lacunae, appeared homologous cell cluster and secreted extracellular matrix that could be stained with Masson stain. In situ hybridization assay showed that chondrocytes expressed of collagen Ⅱ mRNA. It was demonstrated that chondrocytes could secrete collagen Ⅱ with immunohistochemistry method. Conclusion Chondrocytes still secrete collagen Ⅱin three-dimensional culturing system using collagen and human fibrin as supporter. Collagen Ⅱ is one of the most important component of cartilage tissue.

Objective To study the protective effects of Sibelium on neuron injury in rats and its mechanism. Methods The damage model of cultured cortical neuron induced by H₂O₂ was established. The malondialdehyde (MDA) was detected by TBA method, and the breaking rate of DNA was determined by diphenylamine method. The change of transudatory rate of lactate dehydrogenase (LDH), the effects of Sibelium on those indexes described above and its interaction with •OH in vitro were observed. Results The intoxication of H₂O₂ was decreased by Sibelium. Sibelium singnificantly reduced the transudatory rate of LDH, the breaking rate of DNA. The content of MDA in culture and the clearance rate of •OH was increased with the increasing of the dosage of Sibelium. Conclusion Sibelium can obviously protect the cultured cortical neuron against intoxincation of H₂O₂ in rats.

Objective To observe the protective effect of Acanthopanax senticosus saponins (ASS) on myocardial ischemia-reperfusion arrhythmia in rats. Methods The myocardial ischemia-reperfusion model was induced by 30 min coronary occulusion and 60 min reperfusion in open-chest anesthetized rats. The changes of arrhythmia with electrocardiogram lead Ⅱ,the superoxide dismutase (SOD), glutathione peroxidase(GSH-Px) activities , malondialdehyde (MDA) and Ca²⁺ contents in myocardium were determined. Results ASS (25,50, and 100 mg•kg^-1) effectively antagonized the reperfusion injury. It prevented the incidence of reperfusion ventricular arrhythmias (VT and VF), retarded the appearing time and shortened the duration of arrhythmia, lowered the elevated ST segment as well. It decreased significantly the myocardial Ca²⁺ and MDA contents as well as increased the myocardial SOD and GSH-Px activities. Conclusion ASS has protective effect on myocardial ischemia-reperfusion injury, which may be related to scavenging the oxygen free radicals and Ca²⁺ overload formed during reperfusion.

Objective To investigate the effects of ginsenoside Rg₂ on hemodynamics and oxygen metabolism in narco-canine with acute cardiogenic shock. Methods Model of acute cardiogenic shock was made by ligating left anterior descending branch of coronary artery(LAD). Hemodynamics and oxygen metabolism in narco-canine were observed after administrated with 2 and 1 mg•kg^-1 ginsenoside Rg₂. Results Ginsenoside Rg₂ increased mean blood pressure (MBP),left ventricular systolic pressure (LVSP),±dp/dt_max ,and cardiac output (CO), remarkably, and reduced the exponent of oxygen consuming of myocardium and the rate of oxygen intake of myocardium compared with control group. Conclusion Ginsenoside Rg₂ has therapeutical effect on acute cardiogenic shock in canine.

Objective To investigate the effects of chemical H of Renshen Sini Tang on myocardium contraction and action potentials of isolated working heart in adult guinea-pig. Methods Eighteen healthy adult guinea-pigs were randomly divided into control group, low-dose H group and high-dose H group. The ventricular indexes were observed with the methods of perfusion and suspension glass microelectrode on isolated working heart. Results The chemical H of Renshen Sini Tang increased LVSP, LVEDP, ±dp/dt_max, APD₅₀, APA, OS, and V_max. Conclusion The chemical H of Renshen Sini Tang has the effect of increasing myodynamia, and it may activate Na+ and Ca²⁺ channels.

Objective To study the inhibitory effect of 20(S)-ginsenoside Rg₃ on B16 melanoma metastasis and its mechanism. Methods The spontaneous and experimental lung metastasis models of B16 melanoma were used to investigate the inhibitory effects of Rg₃ on tumor metastasis. The assay of tumor-induced angiogenesis was used to observe the effect of ginsenoside Rg₃ on B16 melanoma angiogenesis. The effect of Rg₃ on invasive ability of B16 cells in vitro was detected by Boyden chamber. Results In the spontaneous and experimental lung metastasis models of B16 melanoma, the number of tumor colonies in lung in each Rg₃ group was lower than that in control group. The number of vessels oriented toward the tumor mass and the invasive ability of B16 cells decreased obviously after treated with Rg₃. Conclusion 20(S)-ginsenoside Rg₃ can obviously inhibit the lung metastasis of B16 melanoma. The inhibitory effect partially dues to its inhibiting angiogenesis induced by B16 melanoma and decreasing the invasive ability of B16 cells.

Objective To study the effect of plyrhachis vicina rogher (PVR) on immune function in mice with tumor and the mechanism of tumor inhibition. Methods The BALB/c mice were randomly divided into normal control (NC) group and experimental groups including high, middle and low doses of alcohol extract of PVR. The mice were inoculated with S₁₈₀ cell of ascites sarcoma in mice. The tumor model of animal was established. NC group was fed on distillative water. Experimental groups were fed on 0.5,1.0, and 1.5 g•kg^-1 alcohol extract of PVR for 10 days. The tumor weight,inhibitory rate of tumor,indexes of spleen, IL-2,activities of tumor killer LAK and NK,proliferation of T,B lymphocytes were used as evaluated indexes. Results Spleen weight, killer activities of the LAK and NK, proliferous reaction of the spleen cell and the level of IL-2 secretion in mice treated with alcohol extract of PVR were significantly higher than those in NC group. Conclusion PVR may promote specific and nonspecific immune function of organism and inhibit tumor cells indirectly.

Objective To study the absorpion and utilization of calcium gluconate and calcium lactate in rats in vivo. Methods Weaning male Wistar rats were randomly divided into 5 groups and fed on CaCO₃, skimmed milk powder, calcium gluconate, calcium lactate， and basic low calcium meal in semi-purified diets, respectively, for 4 weeks. Meanwhile，the experiments of 3 days′ feed efficiency and 4 days′ calcium metabolism were carried out on the 20th and 25th day, respectively. Results In calcium-gluconate group, the incremental rate of body weight,the absorption and retention rate of calcium,the femur index, and retention rate of calcium in femur were 92.1%,68.2%,85.1%,4.96, and 4.6%, respectively, all indexes were lower than those in milk-powder group and higher than those in CaCO₃ group (P<0.05 or P<0.01). The items mentioned above in calcium-lactate group were 75.1%,65.5%,82.0%,4.88 and 4.1%, respectively,also higher than those in CaCO₃ group (P<0.05 or P<0.01),no differences were found between calcium-gluconate group and calcium-lactate group except that the incremental rate of body weight in calcium-lactate group was lower than that in calcium-gluconate group.Conclusion Both calcium gluconate and calcium lactate can be served as the good sources for dietary calcium supplement.

Objective To study the effect of microencapsulated tumor cell vaccine against CEA on immune function in tumor-bearing mice. Methods BALB/c mice were inoculated with 1×10⁶ hepatocarcinoma cell line H-22 cells transfected with human CEA encoding sequence. Spleens were removed to prepare single cell suspensions. ConA-induced lymphocyte proliferation and NK cytotoxicity were detected by ³H-TdR incorporation assays. The levels of IL-2 and IFNγ were determined by ELISA and the mRNA expressions of the cytokines were analyzed by RT-PCR. The proportion of T lymphocyte supsets and NK cells were detected by FCM. Results Microencapsulated CEA tumor cell vaccine enhanced mitogen-induced proliferation and NK cytotoxicity of the splenocytes from tumor-bearing mice and promoted the expressions of IL-2 and IFNγ induced with ConA（P<0.05，P<0.01，P<0.05）. Microencapsulated CEA vaccine increased the proportion of CD4 and decreased the proportion of CD8, leading to the increase of CD4/CD8 ratio（P<0.05）. Conclusion Microencapsulated tumor cell vaccine against CEA can promote the immune function in tumor-inoculated mice, indicating that it may be an effective therapeutic vaccine to inhibit CEA positive tumors.

Objective To explore the inhibitory effects of extracts from Trapa manshurica Fler. on liver cancer cells in vivo and in vitro. Methods The sensitivity of extracts from Trapa manshurica Fler. to liver cancer cells in vitro was determined with ³H-TdR incorporation method. The inhibitory rate and growth of the tumor and the weight change of the mice were observed. Results When the dosage was 225.00 g•L^-1, the inhibitory rate of tumor was 75.49% in vitro and the inhibitory rate of tumor was 60% in vivo. Conclusion The extracts from Trapa manshurica Fler. has the inhibitory effects on tumor.

Objective To observe the toxic effects of 2-metheyl-4-chlorophenoxyacetic acid (MCPA) on female mice. Methods The different doses of MCPA (20,100, and 200 mg•kg^-1. Weight.p.o) were given to the mice. The weight,the levels of T₄,TSH, and cholesterol in serum, and the organ coefficient of spleen were analyzed. Results In mid-,high-dose groups, the weight of mice and level of TSH in serum had significantly lowered than that in the control and low-dose groups, but the level of cholesterol in serum was significantly increased than that in low-dose and control groups. In the high-dose group, the level of T₄ in blood was reduced significantly compared with control. Conclusion In the experimental conditions, MCPA can obstruct growing, make endocrine function disorder, and decrease the spleen weight of mice.

Objective To prepare anti-osteosarcoma monoclonal antibodies (McAb) and study its characteristics. Methods Splenocytes of BALB/c mice immunized with osteosarcoma cell line OS-732 were extracted and these cells were fused with murine myeloma cell line NS-1. Enzyme-linked immunosorbent assay (ELISA) and complement-dependent cytotoxic assay were used to screen and three hybriddoma cell lines capable of screting antiosteosarcoma monoclonal antibodies were acquired. The tissues of osteosarcoma, chondrosarcoma, giant cell tumor of bone,leiomyosarcoma, synoviosarcoma, nonossifying fibroma,osteochondroma, normal tissues of muscle and bone were made into frozen slides and the indirect immunofluorescence test were applied to test these specimens. Results All the three lines showed positive results in the osteosarcoma slides and two of them showed weakly positive results in the chondrosarcoma. The three McAb lines combined with different antigenic determinant epitope of osteosarcomous correlative antigens, respectively. Conclusion McAb possesses higher sensitivity and specificity.

Objective To examine the expressions of aquaporin 9(AQP9) in various segments epididymides of adult rats. Methods The capita and caudae epididymides of 12 week rats were removed for total RNA extraction and total protein purification. Northern blotting analysis was performed with AQP9 cDNA probe, and Western blotting was performed with polyclonal anti-AQP9 antibody. Results The expressions of AQP9 were confirmed in rat epididymides and the AQP9 expression in capita epididymides was higher than that in caudae epididymides. Conclusion AQP9 expression is found in rat epididymides. The expression levels of AQP9 in rat capita and caudae epididymides show the reabsorption capacity in different segments of epididymides.

Objective To explore the inhibitory effects of resveratrol (RES) on H²² and MFC in mice. Methods After inoculating tumor cell suspension to oxter of mice, the alternations of tumor growth in mice administrated with RES orally in different doses were observed. Results Inhibitory rates of RES on H²² in different groups (10, 3, and 1 mg• kg^-1) were 53.3%,54.1%, and 47.0%, respectively; inhibitory rates of RES on MFC in different groups (10, 3, and 1 mg• kg^-1) were 64.3%,60.0%, and 47.4%, respectively. Conclusion RES has significant inhibitory effect on H²² and MFC.

Objective To observe the preventive effect of combination fish oil on pathological lesions in inner ear of diabetic rats and its mechanism. Methods Fifty male Wistar rats were randomly divided into three groups randomly :group A (normal control, n=16), group B (diabetic control, n=18), and group C (treatment group,n=16). Rats in group B and group C were injected intravenously with streptozocin(STZ) of 55 mg•kg^-1. Three weeks after injection,the rats in group C were continuously fed on combination fish oil everyday for 9 weeks. The levels of total cholesterol (TC), triglyceride (TG), low density lipoprotein-cholesterol (LDL-C) and very low density lipoprotein-cholesterol (VLDL-C) in blood of rats were detected.The animal auditory brain stem evoked potentials were recorded. Results The levels of TC,TG,LDL, and VLDL in group C descented obviously compared with group B (P<0.05), auditory brain stem evoked potentials wave Ⅲ was obviously shortened in latent period compared with group B (P<0.05) and the threshold of wave Ⅲ decreased (P<0.05). Mass of lipid droplet in hairy cells in inner ear, vacuolar degeneration of mitochondrion and Golgi′s apparatus, hypertrophy and hyperplasia of endothelial cells in micrangium, and thick basement membrane were observed under transmission electron microscopy in group B. But these pathological lesions did not appear in the rats in group C. Conclusion Combination fish oil can prevent the lesions of inner ear in diabetic rats.

Objective To study the inhibitory and repairing effects of deacetylchitosan on gastric ulcer in animal models. Methods Wistar rats were used to set up water-binded stress (n=40), acetic acid-cauterizated (n=32),and gastric mucosa trauma (n=32) models. The rat models were randomly divided into control group (0.5% CMC 10 mL•kg^-1) and experimental groups (Deacetylchitosan Ⅰ： 100 g•L^-1•kg^-1;Deacetylchitosan Ⅱ:50 g•L^-1•kg^-1;Ranitidine:3 g•L^-1•kg^-1). The ulcer indexes and ulcer areas were observed.Results The ulcer indexes in water-binded stress ulcer in Deacetylchitosan Ⅰ, Ⅱ groups and Ranitidine group were lower than that in control （P<0.01），there were no significant differences between experimental groups; The ulcer areas in acetic acid-cauterizated and gastric mucosa trauma ulcer in Deacetylchitosan Ⅰ，Ⅱ groups and Ranitidine group were lower than that in control（P<0.01）, there were no significant differences between experimental groups. Conclusion Deacetylchitosan has inhibitory effects on water-binded stress and acetic acid-cauterizated ulcer, and has repairing effect on gastric mucosa trauma ulcer.

Objective To investigate the effects of valsartan on experimental left ventricular collagen remodeling (LVRM) in rats with myocardial infarction (MI). Methods Ligate anterior descending branches of coronary artery of Sprague-Dawley rats were used to establish models of acute myocardial infarction (AMI). Seventy rats were randomly divided into 6 groups:①Sham groups:including Sham 1(n=5) and Sham 4(n=5).The pericardium of rats in Sham groups were cut open and sutured immediately. The rats were routinely breeded for 1 week and 4 weeks,respectively, 1.5 mL saline was poured into stomach of rats once a day;②Control groups:AMI 1 (n=10) and AMI 4 (n=10). 1.5 mL saline was poured into stomach of rats with AMI once a day for 1 week and 4 weeks,respectively;③Valsartan groups:VAS1 (n=10) and VAS 4 (n=10). Valsartan of the dosage of 10 mg•kg^-1•d^-1 and 1.5 mL saline were poured into stomach of rats with AMI for 1 week and 4 weeks(once a day),respectively. The ratio of collagen type Ⅰ to Ⅲ in non-infarction myocardium in left ventricle was detected by immunohistochemical method. The levels of Ang Ⅱ, aldosterone (ALD), endothelin (ET), thromboxane A₂ (TXA₂), and prostacyclin I₂ (PGI₂) in plasma were measured by radioimmunoassay. Results ①The ratio of collagen type Ⅰ to Ⅲ in non-infarction myocardium in left ventricular was decreased;②Compared with control groups, valsartan could decrease ALD, ET, and TXA₂ and increase AngⅡ and PGI₂ in plasma. Conclusion Valsartan has beneficial effects on LVRM after MI.

Objective To study the detection of gastric cancer micrometastasis in regional lymph nodes by reverse transcriptase polymerase chain reaction (RT-PCR)and its promising application. Methods Tumor tissues of 30 cases of gastric cancer and 126 lymph nodes were studied by both histopathological examination and K19 mRNA RT-PCR. Results K19 mRNA expressed in all specimens of 30 patients with gastric cancer, 26 examined lymph nodes showed postive lymph node metastasis by routine histopathological examination (20.6%), but the number of postive lymph nodes detected by RT-PCR increased to 42 (33.3%). After K19 and β-actin were amplificated by RT-PCR, the amplification fragments of 460 and 540 bp appeared in all cancer and gastrointestinal tissues. Conclusion K19mRNA detection by RT-PCR is superior to routine histpathological examination and is a highly sensitive and specific method.

Objective To explore the expressions of tumor-related genes (MMP-2 and PTEN) in colorectal carcinoma tissues and their clinic pathological significances. Methods The expressions of MMP-2 and PTEN in 40 patients with colorectal carcinoma were investigated by RT-PCR. Results ①There existed significant differences of the expressions of MMP-2 and PTEN between normal control and Dukes A,B (P<0.01); as well as the expressions of MMP-2 and PTEN between Dukes C,D and Dukes A,B(P<0.05); ②There existed significant differences of the expression rates of MMP-2 and PTEN between the high or middle differentiation grade of the carcinoma and the low differentiation grade (P<0.05);③There existed significant differences of the expression rates of MMP-2 and PTEN between the patients with lymph metastasis and those without lymph metastasis (P<0.05). Conclusion The expressions of MMP-2 and PTEN have close relationship with the pathology of colorectal carcinoma and their own biology behaviors. Both MMP-2 and PTEN play important roles in distincting hyperplasia of colorectal carcinoma.

Objective To examine the changes of T lymphocyte subsets, IL-10 and IL-12 levels in patients with chronic hepatic diseases. Methods Serum IL-10, IL-12 and T lymphocyte subsets in 68 patients with various clinic types of viral hepatic diseases were detected by double-sandwich ELISA and erythrocyte garland test, respectively. Results CD4⁺，CD4⁺/CD8⁺ in patients with different clinic types of chronic hepatic disease were significantly decreased, and CD8⁺ cells were significantly increased compared with control, and IL-10, IL-12 levels in patients with chronic hepatitis B and hepatocirrhosis markedly increased. CD4⁺, CD4⁺/CD8⁺, and IL-12 in HBeAg positive hepatitis group were markedly lower than those in HBeAg negative hepatitis group. The number of CD8⁺ cells and IL-10 level were significantly higher than those in HBeAg negative hepatitis group. Conclusion The immunological function is disorder in patients with chronic HBV hepatic disease, and CD4⁺, CD4⁺/ CD8⁺, IL-10, and IL-12 levels are related to state of HBV replication carrier or the active degree of HBV in patients with chronic hepatic disease.

Objective To compare the differences of creep between heavy-load articular cartilage and non-heavy-load articular cartilage,and to discover the correlation between creep and biochemical compound. Methods The creep effects of normal humeral head cartilage, femoral cartilage and femoral condyle cartilage of male healthy ptoma were measured by testing on the AG-10TA automatic control electrical all-purpose experimental machine. Results The articular cartilage creep experimental data in three groups were analyzed, the value of heavy-load artilcular cartilage was higher than that of non-heavy-load one. The strain-time curves and functions under even stress were obtained. Conclusion Heavy-load articular cartilage has better creep effects than non-heavy-load one. Furthermore, the creep effects of articular cartilage are interrelated with its biochemical compound.

Objective To evaluate the effects of hepatocyte growth factor(HGF) on intravasation migration of cancer cell. Methods The model of intravasation migration of cancer cell in vitro was used. The fibrosarcoma cell HT1080 was labelled with fluorescence, and the fluorescence guantities in 24-pore plate through collagen membrane and HUVEC single cells layer were detected in different time to eveluate the intravasation migration of cancer cell.Results 12,24, and 48 hours after administration,the fluorescence quantity in experimental group was significantly higher than that in control group (P<0.01). Conclusion HGF can promote the intravasation migration of cancer cells.

Objective To investigate Flt₃ tandem duplication mutation (ITD) and its distribution in different malignant hematopoietic diseases. Methods PCR was used to detect the Flt₃-ITD mutation in blast cell of bone marrow from 103 patients with malignant hematopoietic disease. Results Flt₃-ITD mutations were found in 18.2% （8/44）of the patients with primary acute myeloid leukemia (AML), more Flt₃ aberrations were found in patients with FAB M1. Flt₃-ITD mutations were not found in patients with acute lymphoid leukemia (ALL), myelodysplastic syndrome(MDS), multiple myeloma(MM), non-Hodgkin′s lymphoma(NHL), and chronic myeloid leukemia (CML). Conclusion Flt₃-ITD can be a important molecular marker for AML.

Objective To study the value of adhesion molecules in the mechanism of peripheral blood stem cell mobilization. Methods Flow cytometry was used to detect the expressions of adhesion molecules of very late antigen 4 (VLA-4)，leukocyte function-associated antigen 1 (LFA-1) and L-selectin (CD62L) on CD34⁺cells from granulocyte colony-stimulating factor (G-CSF)-mobilized peripheral blood(PB) and bone marrow(BM). Results The expressions of VLA-4 and LFA-1 were decreased significantly on CD34⁺cells from G-CSF-mobilized peripheral blood than those from bone marrow, and the expression of CD62L showed no significant difference. Conclusion The reduction of adhesion molecules of VLA-4 and LFA-1 may be an important step during the course of G-CSF-mobilized hematopoietic stem cell into peripheral blood.

Objective To study the expression of epithelial-cadherin (E-cd) in breast cancer cells and the microvessel density (MVD) in breast cancer mesenchyma, and to analyze the correlation between two factors and metastasis in breast cancer. Methods Seventy breast cancer cases with or without lymph node metastasis were selected. The routine histopathology was used to make pathological diagnosis and histological type. Immunohistochemical method was applied to detect the expressions of E-cd and CD34. Results The expression of E-cd was relative to the lymph node metastasis (P<0.05) and irrelevant to the histological types of breast cancer. The MVD in breast cancer with lymph node metastasis was significantly higher than that without lymph node metastasis, and also was irrelevant to the histological types of breast cancer. When the grade of the MVD in breast cancer mesenchyma was low, there was no difference of MVD between lymph node with metastasis and without metastasis(P>0.5),but it was relative to the MVD of lymph node(P<0.001). Conclusion Breast cancer metastasis is dependent on not only the E-cd gene mutation but also the mesenchymal MVD. Therefore, the expression of E-cd in breast cancer cells and the examination of MVD can be used as a reference marker to diagnose the breast cancer metastasis.

Objective To investigate the clinical diagnostic value of bone marrow prostate specific antigen (PSA) in bone metastasis of prostatic cancer. Methods Twenty cases of prostatic cancer were divided into positive bone scanning group (n=12) and negative bone scanning group (n=8) according to the results of isotope bone scanning,and 20 cases of benign prostatic hyperplasia were used as control.The levels of PSA in bone marrow and serum were measured in all patients to study the correlationship of bone marrow PSA and bone metastasis, and the ratio of bone marrow PSA to serum PSA was calculated. Results The level of PSA in bone marrow in positive bone scanning group were statistically higher than those in negative bone scanning group and control group (P<0.05)). The diagnostic ratio of bone marrow PSA to serum PSA had been set to 0.70, the sensitivity to predict bone metastasis was 78%, the specificity was 86%, the accuracy was 73%. Conclusion The level of PSA in bone marrow has close correlation with bone metastasis of prostatic cancer and it is a marker of bone metastasis of prostate cancer. The ratio of 0.70 of bone marrow PSA to serum PSA can be used as a index to diagnose bone metastasis.

Objective To study the relation between bcl-2，proliferating cell nuclear antigen (PCNA) and endometrium and endometriosis. Methods The expressions of Bcl-2 and PCNA in eutopic endometrium from 46 cases during the menstrual cycle and ectopic endometrium from 15 patients with endometriosis were examined by immunohistochemical mothod. Results The staining of Bcl-2 protein on cytoplasm of glandular cells was mainly seen in proliferative phase. It rarely appeared in secretory phase and absent during menstruation. It appeared highly in endometriosis. Conclusion The expressions of Bcl-2 protein and PCNA protein in eutopic endometrium during the menstrual cycle have periodical changes ,and differ from that in endometriosis.

Objective To study the pre-emptive analgesia effects of ketamine on postoperative pain management and stress responses. Methods Forty-five patients with hysteromyoma undergone hysteromyomectomy were randomly assigned to three groups (n=15，each group): group Ⅰ, control group without pre-emptive analgesia; group Ⅱ, the patients were administrated with 0.2 mg•kg^-1 ketamine intravenously 30 min before operation; group Ⅲ, 30 mg ketamine was administrated into epidural analgesia 30 min before operation. All patients received postoperative analgesia with patient controlled epidural analgesia（PCEA）. The VAS and first PCEA trigger time (min), morphine consumption (mg) and the number of PCEA successful triggers, analgesic-related adverse effects were observed. The changes of epinephrine (E), norepinephrine (NE) in plasma were detected by improved fluorescence method, and cortisol level in serum were also measured by radioimmunoassay. Results The VAS were significantly different between groupⅠ,Ⅱand Ⅲ. The first PCEA trigger times of groupⅡ and Ⅲ were longer than that of groupⅠ. The number of PCEA successful triggers and morphine consumptions two days after operation in group Ⅱ and Ⅲ were significantly less than those in groupⅠ; the levels of E and NE in plasma were increased in group Ⅱ and Ⅲ one day after operation and in group Ⅰ during 48 hours after operation, but the degree in group Ⅱ and Ⅲ was lower than that in groupⅠtwo days after operation. Conclusion The pre-emptive analgesia of ketamine can decrease morphine consumption, stress response time and analgesic-related adverse effects.

Objective To determine the changes of cytokines and inflammatory mediators in systemic inflammatory response syndrome (SIRS) in acute abdominal disease. Methods The fasting peripheral venous blood were collected in 35 patients with SIRS in acute abdominal diseases on the day of admission,and 1,3,7, and 14 days after admission, respectively. The levels of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) were detected by enzyme linked immunosorbent assay. The level of endotoxin in plasma was determined by tripetide azo-colorimetric method,the levels of nitric oxide (NO) and lipid peroxide (LPO) in serum were examined by fluorospectro photometry. Results The levels of TNF-α, IL-6, and endotoxin in patients were obviously higher than those in control group (P<0.01); IL-6 and TNF-α levels peaked on admission, then decreased gradually. IL-6 decreased obviously on the third and seventh days; endotoxin maintained higher level within the first 3 days on admission, but obviously lowered on the seventh and fourteenth days; serum LPO levels were high on admission, the third and seventh days (P<0.01), so were the levels of NO on admission and the third day, (P<0.05); NO and LPO maintained relatively higher levels. Conclusion There is usually infectious SIRS in acute abdominal diseases; the levels IL-6, TNF-α, NO, LPO, and endotoxin are increased in SIRS.

Objective To study the consistency of the results between of MR myocardial perfusion imaging and DSA coronary arteriography. Methods MR myocardial perfusion imaging and DSA cornary artery imaging were perfromed to 51 patients with coronary heart disease with typical clinical symptoms and typical cardiogram. Dynamic contrast enhanced myocardial imaging (DCEMI) myocardial perfusion imaging analysis software for GE AW4.0 working station was employed to analyze the first pass phase images of the myocardial perfusion of the left ventricle short axle. A curve of the time-intensity of the cardiac muscle perfusion was obtained and the rising slope and the peak value of signal intensity were observed. According to the description of the relation between the position of the short axle of left ventricle and the distribution of the coronary artery blood vessels, the existence of narrowed coronary artery was determined. DSA machine of DAR 1 200 double C arm was employed in the coronary artery imaging. Judkin method employing Seldinger′s method of femoral centesis was carried out and the value ≤50% of the diameter of the coronary artery was determined as the diagnostic standard of narrowness of clinical significance. Results The results of the two methods in 43 cases were consistent and 8 not, the rate of consistency was 84.3%. Conclusion High consistency of the results was found between MR myocardial perfusion imaging and DSA coronary arteriography. MR myocardial perfusion imaging is a non-traumatic method to determine the blood flow of myocardium and it can supply valuable information and guidance for the diagnosis and treatment for the patients.

Objective To evaluate the children′s neurobehavioral functions after comprehensive management of Songhua River by mercury. Methods The polluted region by mercury of Songhua River after comprehensive management of Jilin City was used as survey group, and unpolluted region by mercury of Changchun City as control group. The mercury levels in water, soil, crops, and fish were detected in the two groups. Neurobehavioral Core Test Battery (NCTB) recommended by WHO was used to examine the neurobehavioral functions of 127 children in survey group and 156 children in control group. Results There were no significant differences of mercury content in river water, soil, and fish between survey and control groups, but the mercury content in crops of survey group was significantly higher than that of control group (P<0.05), and it was lower than that of national standards. There were no significant differences in six neurobehavioral functions between survey and control groups (P>0.05), only the wrong score of Pursuit Aiming Test Ⅱ in the survey group children was larger than that in the control group notably (P<0.01). Conclusion The comprehensive management of mercury pollution source has resulted in great improvement of environment quality, which has no significant effects on the children′s neurobehavioral functions.

Objective To analyze the effects of some routine reagents of coliform protein on two kinds of protein qualification (dyebinding method and bicinchoninic acid method) was used.Methods Two way analysis of variance (two way ANOVA) was used. Results Bicinchoninic acid method was interfered by the reagents containing EDTA or glucose（P<0.05）. No interference to the method was shown by using the reagents containing guanidine-HCl or urea. As for dyebinding method, all above reagents had not interference effects. Conclusion The interference of reagents can not be neglected in protein qualification.

Objective To analyze mitochondrial DNA polymorphism of district of Lubunour at the Bronze Age in Xinjiang. Methods MtDNA was extracted from 10 ancient individual bones at the tombs of Lubunour. Through four overlapping primers, mtDNA HVS-1 was amplified and sequenced. Results 10 nucleotide sequences of 363 bp length were obtained. Phylogenetic analysis indicated that the nucleotide diversity and mean pairwise differences of ancient people in Lubunour was close to European population. Conclusion An ancient European population had existed prior to 3800 years ago.
