加味四逆散对身心应激模型大鼠胃肠功能及胃组织中GASR和空肠组织中VIPR2 mRNA表达的影响

Effects of Jiaweisinisan on gastrointestinal function and expressions of GASR mRNA in stomach tissue and VIPR2 mRNA in jejunal tissue of psychological stress model rats

摘要: 目的:建立大鼠慢性身心应激模型,探讨加味四逆散(JWSNS)对模型大鼠胃排空率及小肠推进比、大鼠下丘脑环核苷酸系统(cAMP、cGMP)、胃组织中胃泌素受体(GASR)与空肠组织中血管活性肠肽Ⅱ型受体(VIPR2)mRNA表达的影响,阐明JWSNS干预应激性胃肠功能障碍的机制。方法:60只大鼠随机分为正常组,模型组,小、中、大剂量JWSNS组和西沙比利组,每组10只。采用慢性心理性应激方法建立大鼠应激模型,经JWSNS等干预后检测胃肠动力,放射免疫法检测大鼠下丘脑中cAMP和cGMP含量,RT-PCR法检测胃组织中GASR及空肠组织中VIPR2 mRNA表达变化。结果:与模型组比较,各治疗组大鼠下丘脑中cAMP和cGMP 含量显降低(P<0.05或P<0.01);与西沙比利组比较,大剂量JWSNS组大鼠下丘脑cAMP和cGMP 含量显著降低(P<0.05或P<0.01)。与模型组比较,各治疗组胃排空率及小肠推进比均明显升高(P<0.05或P<0.01);与西沙比利组比较,大剂量JWSNS组胃排空率升高不明显(P>0.05),而小肠推进比明显升高 (P<0.05)。各治疗组大鼠胃组织中GASR mRNA表达水平均高于模型组,空肠组织中VIPR2 mRNA表达水平则降低(P<0.05或P<0.01);中和大剂量JWSNS组大鼠胃组织中GASR mRNA表达水平均高于西沙比利组 (P<0.05或P<0.01);大剂量JWSNS组大鼠空肠组织中VIPR2 mRNA表达水平低于西沙比利组 (P<0.01)。结论:环核苷酸系统(cAMP、cGMP)异常可影响胃肠功能,JWSNS可通过影响环核苷酸系统对慢性心理应激反应进行调控,进而改善胃肠功能,并可改善胃组织中GASR和空肠组织中VIPR2 mRNA的表达。

Abstract: To establish  rat models of  chronic psychological stress and to study the effects of Jiaweisinisan(JWSNS) on gastric emptying rate and small intestinal propulsion ratio,cyclic nucleotide system in rat hypothalamus ( cAMP,CGMP ),and mRNA expressions of gastrin receptor (GASR) in stomach tissue and vasoactive intestinal peptide II receptor ( VIPR2) in jejunal tissue of rat models,and to elucidate the mechanism of JWSNS in effecting stress-induced gastrointestinal dysfunction. Methods 60 rats were randomly divided into normal group,model group,small,middle,big doses of JWSNS  groups,and Xishabilly group,10 in each group.The rat models were set up by chronic psychological stress method,after JWSNS intervention,the gastrointestinal motility was detected,and radioimmunoassay was used to detect the changes of cAMP and cGMPcontents in hypothalamus of rats,and the mRNA expression changes of GASR in stomach tissue and VIPR2 in jejunal tissue of rats were measured with RT-PCR method.
Results  Compared with model group,the contents of hypothalamus cGMP and cAMP in treatment groups were decreased,there were  significant differences (P<0.05or P<0.01);compared with Xishabilly group,the contents of hypothalamus cGMP and cAMP in big dose of JWSNS  group were decreased,there were significant differences(P<0.05 or P<0.01).Compared with model group,the gastric emptying rate and small intestinal propulsion ratio in treatment groups were significantly increased (P<0.05 or P<0
.01);compared with Xishabilly group,the gastric emptying rate in  big dose of JWSNS group was not significantly increased (P>0.05),and the small intestinal propulsion ratio was increased significantly (P<0.05). Compared with model group,the GASR mRNA expression levels in stomach tissue in treatment groups were increased,and the VIPR2 mRNA expression levels in jejunal tissue were decreased,the differences were statistically significant (P<0.05 or P<0.01);the GASR mRNA expression levels in middle   and big doses of JWSNS  groups were higher  than that in Xishabilly group (P<0.05 or P<0.01),and the VIPR2 mRNA expression level in jejunal tissue in  big dose of JWSNS group was lower than that in  Xishabilly group ( P<0.01).
Conclusion The cyclic nucleotide system (cAMP,CGMP) abnormalities can affect the gastrointestinal function and JWSNS can regulate chronic psychological stress response by influencing cyclic nucleotide system,thereby improve the gastrointestinal function,and improve the expressions of VIPR2 and GASR mRNA in stomach and jejunum tissues.
