龙牙葱木皂苷对实验性心力衰竭大鼠心功能的影响

摘要: 目的:观察龙牙葱木皂苷(Aralosides,As)对实验性心力衰竭大鼠血流动力学的影响。 方法:Wistar大鼠24只,静脉注射1.5%戊巴比妥钠溶液成功建立心力衰竭模型平稳5 min后,阴性对照组、As实验组及阳性药物对照组各8只大鼠,分别静脉注射生理盐水(0.9 mg•kg-1)、龙牙葱木皂苷(As,10 mg•kg-1)和去乙酰毛花甙(0.04 mg•kg-1),各组均以心力衰竭模型后5 min时的各项心功能指标为对照,采用四导生理记录仪同步记录正常时、心衰后5 min以及静脉给药后30 min内各组大鼠的左室收缩压(LVSP)、左室最大上升和下降速率(±dp/dtmax)、左室舒末压(LVEDP)、收缩压(SAP)、舒张压(DAP)和心率(HR)等心功能指标,计算机监视记录结果及分析,用彩色绘图仪绘制曲线。 结果:心力衰竭模型大鼠静脉注射As后,LVSP由(10.8±1.0) kPa增加到(14.5±1.8) kPa (P<0.001),+dp/dtmax由(163±24)kPa•s-1增加到(295±66) kPa•s-1 (P<0.001),-dp/dtmax由(-140±31) kPa•s-1下降到(-218±60)kPa•s-1(P<0.01),LVEDP由(1.46±0.24)kPa下降到(1.28±0.17)kPa(P<0.05);静脉注射去乙酰毛花甙后,LVSP由(10.5±1.2)kPa增加到(12.9±1.6)kPa(P<0.01),+dp/dtmax由(144±26)kPa•s-1增加到(222±53)kPa•s-1(P<0.001),-dp/dtmax由(-132±23) kPa•s-1下降到(-190±32)kPa•s-1 (P<0.001),LVEDP由(1.31±0.19)kPa可下降到(1.14±0.18)kPa(P<0.05);而心力衰竭大鼠静脉注射生理盐水后,其心功能的各项指标较注射生理盐水前均无明显改变(P>0.05)。结论:As具有与去乙酰毛花甙一致的抗心力衰竭。

Abstract: Objective To study the effects of Aralosides (As) on cardiac function in rats with experimental heart failure. Methods Twenty-four experimental heart failure rat models induced by 1.5% pentobarbital sodium were researched by using hemodynamic method and then were devided into control group(salt solution, 0.9 mg•kg-1), As group(As, 10 mg•kg-1) and deslanoside group (deslanoside, 0.04 mg•kg-1). Every group was controlled with the hemodynamic value 5 minutes after heart failure. Left ventricular systolic pressure (LVSP), left ventricular end diastolic pressure (LVEDP), maximal rising and falling rate of ventricular pressure(±dp/dtmax) were recorded with 4-channel polygraph to observe the the effects of salt solution, Aralosides and deslanoside on cardiac function in rats with experimental heart failure. Results In As group, the LVSP was increased from (10.8±1.0) kPa to (14.5±1.8)kPa (P<0.001),+dp/dtmax was increased from (163±24)kPa•s-1 to (295±66)kPa•s-1 (P<0.001),-dp/dtmax was decreased from (-140±31) kPa•s-1 to (-218 ±60) kPa•s-1 (P<0.01) and LVEDP was decreased from (1.46±0.24) kPa to (1.28±0.17) kPa (P<0.05) after intravenous administration of As. In deslanoside group, the LVSP was increased from (10.5±1.2) kPa to (12.9±1.6) kPa (P<0.01),+dp/dtmax was increased from (144±26) kPa•s-1 to (222±53) kPa•s-1 (P<0.001),-dp/dtmax was decreased from (-132±23) kPa•s-1 to (-190±32)kPa•s-1(P<0.001) ,and LVEDP was decreased from (1.31±0.19) kPa to (1.14±0.18)kPa (P<0.05) after intravenous adminstration of deslanoside. But the hemodynamic value did not change after intravenous administration of salt solution (P>0.05). Conclusion As can induce a positive inotropic action on heart failure myocardium, performing in a similar manner as deslanoside.
