生长抑素和内皮抑素对小鼠肿瘤血管新生的抑制作用

Inhibitory effects of angiostatin and endostatin on angiogenesis in mice

摘要: 目的:直视观察小鼠背部开窗模型肿瘤生长情况及测定内皮生长因子、抗原CD34的表达,了解血管生长抑素和内皮抑素对肿瘤的作用。 方法:本实验设对照组、实验对照组及低、高剂量实验组,建立小鼠背部开窗模型,分别给低、高剂量血管生长抑素及内皮抑素(AS 0.6 ng•kg-1+ES 2.5 mg•kg-1和AS 1.2 ng•kg-1+ES 5.0 mg•kg-1),在13 d高剂量实验组肿瘤生长受抑制情况明显,13 d后处死,采用免疫组织化学方法测定内皮生长因子及抗原CD34的表达,了解肿瘤血管新生受抑制情况。 结果:肿瘤细胞移植后, 每天直视下观察肿瘤生长情况,对照组所有移植肿瘤生长状态良好, 肿瘤长大使小鼠行动发生困难;给药组较对照组肿瘤生长慢且小, 行动不受限制,肿瘤的生长被明显抑制。在对照组中内皮生长因子和抗原CD34染色高表达, 在给药组则低表达。 结论:血管生长抑素和内皮抑素在抑制肿瘤血管新生中起重要作用。

Abstract: Objective Using skylight tumor model to observe the tumor growth and effects of angiostatin(AS) and endostatin(ES) on tumor. Methods The mice were randomly divided into four groups: blank contrast group, experimental contrast group, low dose experimental group(AS 0.6 ng•kg-1+ES:2.5 mg•kg-1), and high dose experimental group(AS 1.2 ng•kg-1+ES 5.0 mg•kg-1). Mice skylight tumor models were established.After 13 days, the tumor growth was prohibited obviously observed with the naked eye in high dose experimental group and the expressions of CD34 and VEGF were determined with immunohistochemistry to explore mechanism in the course of anti- angiogenesis. Results After the tumor cells had been transplanted into mice, it was difficult for the mice to go in experimental contrast group. But the tumor growth in drug experimental groups were slow and the mice could slow and walk well. The expressions of VEGF and CD34 were higher in experimental contrast group than those in drug experimental groups. Conclusion AS and ES can have important inhibitory effects on angiogenesis.
