复合培养人工皮肤的细胞生物学规律

Cell biology regularity of double layer cultured artificial skin

摘要: 目的:探讨复合培养人工皮肤构建过程中的细胞生物学规律。 方法:应用组织工程学技术制备人工皮肤,计算收缩指数,3H-TdR掺入法测定DNA合 成能力,3H-脯氨酸掺入法测定培养人工皮肤胶原蛋白合成能力。 结果:培养12 d之内,人工皮肤的收缩指数只有5%左右;14 d后开始出现较大的同心圆收缩,收缩指数14%~27%;23 d后收缩指数维持在30%左右。3H-TdR掺入率及3H-脯氨酸掺入率随着培养时间的延长而逐渐升高。 结论:复合培养人工皮肤增殖活跃,收缩不明显,具有合成细胞外基质的能力。

Abstract: Objective To study the cell biology regularity of double layer cultured artificial skin. Methods The double layer cultured artificial skin of rat was reconstructed by using tissue engineering technology. The contractile indexes of the double layer cultured artificial skin were detected everyday, the synthesis ability of DNA was evaluated by 3H-TdR incorporation method and the synthesis ability of collagen protein was evaluated by 3H-proline method. Results Within 12 days of cultureng, the contractile index of cultured artificial skin was about 5%. After the 14th day, the contractile index of cultured artificiaskin was among 14%-27%. After the 23th day of culturing, the contractile index of cultured artificial skin was retained about 30%. The incorporation efficiencies of 3H-TdR and 3H-proline were increaced gradually. Conclusion The double layer cultured artificial skin has low ability of contractile and high abilities of proliferation and extracellular matrix synthesis.
