牛视网膜毛细血管周细胞的原代培养

Primary culture of bovine retinal capillary pericytes

摘要: 目的:建立牛视网膜毛细血管周细胞原代培养的方法。 方法:以酶消化法原代培养牛视网膜毛细血管周细胞,用透射电镜和免疫细胞化学法对细胞 进行鉴定。 结果:培养的周细胞贴壁生长,胞体肥大,呈不规则状、有较多突起。细胞生长缓慢,呈现非接触抑制的重叠状生长。电镜下周细胞核大、不规则、有切迹,可见多个核仁。细胞表面有微绒毛,胞质内有丰富的细胞器,无Ⅷ因子小体;免疫细胞化学显示细胞呈平滑肌肌动蛋白(α-SMA)阳性,Ⅷ因子相关抗原阴性。 结论:酶消化法原代培养牛视网膜周细胞是一种稳定、可靠的方法。

Abstract: Objective To establish a method for the primary culture of bovine retinal capillary pericytes. Methods Bovine retinal capillary pericytes were cultured in vitro with enzyme digestion method. Electron microscopy and immunocytochemical method were used to evaluate the cultivated cells. Results The cells attached to the wall of culture dishes while growing and showed hypertrophy, irregular shape with more apophysis. Big, irregular nucleus with multiple nucleoli could be seen in the cytoplasm by transmission electron microscopy. The cultivated cells showed positive reaction for α-smooth muscle actin (α-SMA) antibody, and negative reaction for factor Ⅷ antibody with immunocytochemical assay. Conclusion The enzyme digestion procedure is a stable and reliable method to obtain bovine retinal pericytes.
